Lilium spp.is a perennial herb belonging to Lilium family,a subclass of monocotyledons.It is one of the five largest fresh cut flowers in the world.It has very high ornamental value and economic value.At present,Lily provenances still depend on imported seed balls,which are expensive,especially Oriental Lily series.On the other hand,the traditional propagation methods of Lily are inefficient,virus infection,variety degradation and other serious problems,which seriously affect the quality of lily.Therefore,rapid propagation of a large number of lily bulbs is an important task for our research.In recent years,in vitro culture of different lilies has been carried out in China,but there are many varieties of lilies with great differences.In order to lay a foundation for genetic transformation,virus detoxification,tissue rapid propagation and industrialized production of Oriental lily,Sorbonne was selected as the research object,and its scales and leaves were used as explants to explore the efficient regeneration system of Oriental lily.The main results are as follows:In the study of establishing graft regeneration system,the best disinfection methods were 75%alcohol for 30 seconds,sterile water for one time,sodium hypochlorite for 20 minutes,and sterile water for five times.The pollution rate of this method is the lowest,16.67%.The best medium for inducing bulblets from scales and leaves was MS+2.0 mg/L 6-BA+0.2 mg/L NAA,MS+0.5 mg/L 6-BA+0.2 mg/L NAA,respectively.The best medium for bulblet proliferation was MS+2.0 mg/L 6-BA+0.2 mg/L NAA,and the multiplication coefficient was 2.46.The addition of 30 g/L sucrose concentration to the medium induced the best effect of bulb expansion.The best medium for rooting of aseptic seedlings was 1/2MS,and NAA was not conducive to rooting.The best transplanting matrix formula is peat soil:vermiculite is 1:3.In the study of establishing indirect regeneration system,the best medium for inducing callus from leaves of oriental lily is ’Sorbonne’ was MS+2.0 mg/L Pic,with the induction rate as high as 94.00%.Suspension culture could effectively proliferate callus,improve the state of callus and enhance the regeneration ability of callus.The optimum medium for callus differentiation of Sorbonne was MS+0.5 mg/L NAA.The optimum concentration of hygromycin was 20 mg/L.
