Worldwide, rice blast caused by the filamentous fungus Magnaporthe grisea is one of the most economically devastating crop diseases, Management of rice blast via breeding blast-resistant varieties has had only short-term success due to the frequent breakdown of resistance under field conditions. So the researchers attach importance to utilize horizontal resistant cultivars increasingly. We choice "Minghui63" as research materials, which was the high resistence cultivar in 1980s in China, We research the difference expression proteins of sheath using 2-DE technology after infection by M.grisea in virto for the seak of discovering differential resistance-related proteins and better know the mechanism of rice blast.The different expression proteins on the sheath inoculated by the virulence and avirulence rice blast fungus in virto were analyzed by 2-dimensional electrophoresitic technology. Many protein spots resolved on 2-D gels were found to be differentially expressed after analyzing silver stained gels of the soluble and membrane protein samples from the control and 24 hours ,48 hours,96hours post inoculation. we have got the following results.1. The accordant different expression proteins of sheath after inoculated by pathogenic and non pathogenic rice blast fungi M.grisea in virto.We have found 17 spots in all on the gels of the sample after inoculated by M.grisea for 24hours in virto.but there are only 5 proteins express either inoculated by pathogenic or non pathogenic rice blast fungi. After inoculated for 48 hours,the same express proteins is 11 of 23,and 96 hours is 6 of 27,These data show that there are so many distinctness after inoculated for 24 hours between two kinds of reactions,and 96 hours is the same like 24 hours,but after inoculated for 48 hours the are more common ground between two reactions.2.The differential different expression proteins after inoculated by pathogenic and non pathogenic rice blast fungi M.grisea in virto.Analyzing each period of time after inoculated by M.grisea in virto shows that non pathogenic rice blast fungi in a position to induce more proteins, but there are more and more differential proteins expression induced by pathogenic rice blast fungi along with time. There are 39.3% upgrade proteins induced by pathogenic rice blast fungi, and the non pathogenic is 64.3%.This indicates different kinds of reaction hold different resistant mechanism.3. Compare the different expression proteins lengthwaysCompare all of the different expression proteins lengthways shows that not noly the amount of the proteins induced by non pathogenic rice blast fungi are more but also earlier than the proteins induced by pathogenic rice blast fungi. These results accord with the other researchers.4. The significance of this work and the future workIn the future work, we will analysis these proteins by MS or other methods right along to know more detailed information,and it will redound to better know the mechanism of rice blast,Providing theoretics gist for resistence inducement and so on.
