Detection And Sequence Analisis Of Necleoprotein(N) Gene Of Rabies Viruses In Animal From Guangxi

In this study, 1278 brain samples of domestic dogs from different areas in Guangxiprovince were collected and detected by RT-PCR, and 35 samples were positive. Thepositive rate of samples from rabies dogs and healthy dogs were 100% (11/11) and 1.89%(24/1267) respectively. Being opposite to the epidemic situation of human rabies inGuangxi, samples of healthy dogs from severe epidemic areas such as Fangchenggangand Baise were all negative. Moreover, brain samples of 3 cattles and 1 pig sufferingrabies and 1 normal cat were also positive detected by RT-PCR.Then, 460-bp fragment of N genes of 25 above-mentioned rabies isolates werecloned and sequenced. The results of homology comparison and phylogenetic analysisindicated that all rabies virus isolated from Guangxi are genotype I of lyssavirus and couldbe divided intoâ… ,â…¡,â…¢andâ…£4 sublineages There are 15 isolates, includingGXGL1, GXLZ1, GXYL1, GXGL2, GXGG1, GXNN1, GXNN5, GXWZ1, GXQZ1,GX260, GXCH3, GXCH1, GXCH2, GXNN3, GXNN9, belong to sublineageâ… , theirnucleotide homologies were 97.6-100%. Only one isolate GXNN6 belongs to sublineageâ…¡:its nucleotide homology of nucleoprotein gene with 8738THA was 95.8%. 5 isolatesGXCH7, GXBS1,, GXHC1, GXGG2, GXQZ4 composed sublineageâ…¢, theirnucleotide homologies are 98.5-100%.There are 4 isolates belong to sublineageâ…£,including GXYL3, GXCH4, GXYL2, GXBH1, their nucleotide homologies ofnucleoprotein gene with ERA are 99.1-99.6%. In addition, phylogenetic datademonstrated the distributions of above-mentioned 4 sublineages had some geographicalcharacter, sublineageâ… mainly distribute in the east region of Guangxi, sublineageâ…¡hasonly one strain GXNN6 which isolated from Nanning and it is in same sublineage withthose strains isolated from Thailand and Malaya.
