Object:Recently,the rabies of Guangxi Province is highest in China,and the dog is the rabies-carrying host primarily.In thid study,The research of the prevalence and the extent of genetic diversity of rabies in Guangxi should be useful for prevention and control of the rabies.Method:485 samples of brain and 387 samples of saliva were collected from 12 cities of Guangxi and detected by reverse transcription-polymerase chain reaction(RT-PCR),then the positive samples were inoculated to mice cerebrally.Cloned and sequenced of Nucleoprotein(N)and Glycoprotein(G) genes,and the sequences were analyzed by DNASTAR.Result:There are 11 positive samples isolated from brain,1 sample is isolated from one positive dog' saliva.The phylogennetic tree show the 11 isolates belong toⅠ,Ⅱgroups.GXNND(T),GXHXB,GXLA11 and GXQZD belong to groupⅠ,and the GXLC9,GXPL,GXLCC,GXYZD,GXLA11, GXNN2,GXPXD belong to groupⅡ.The homology of Nucleoprotein genes between strains is 89.0%-99.9%for the nucleic acid,97.8%-99.8%at the amino acids level,it can found the main amino acids mutations at 90,110,332 and 397 in nucleoprotein groupⅠ,but at 42 and 332 in nucleoprotein groupⅡ,these amino acid mutations have specificity of group.The homology of Glycoprotein genes between straits is 86.9%-99.9%for the nucleic acid,93.9%-99.8%at the amino acids level,the signal peptide and mature peptide of Glycoprotein were found to be high variable,the main amino acids mutations were at -6,-7,96,132, 156,436,443,and 447 in Glycoprotein groupⅠ,but -4,-5,-15,-18,90,156, 168,204,249,253,289,332,382,427,443,463,474,486 and 487 in Glycoprotein groupⅡ.They also have specificity of group.The antigen epitope of the N P are not mutated,and the phosphorylation Ser389 in groupⅠ,too.the Helper T cell(Th)epitope are also conservative.The 37aa and 319aa are the glycosylation sites of the G P,and the GⅠ,GⅡare not mutated,but the GⅢin groupⅡis mutated:Val332→Ile332.Most of the isolates are from Nanning and Liuzhou of Guangxi,the isolates of groupⅡare mainly found in the western region of Guangxi,but the isolates of groupⅠare mainly found in the eastern region of Guangxi.
