Microsatellite or Simple Sequence Repeat is a newly appeared molecular marker.Because of its stabilization, high polymorphism, and co-dominate, it was used widely byinvestigators. Because less research was done about plum microsatellite, following investigations were done in this paper:1. We obtained 61 microsatellite sequences from enriched fragments by magnesphere,ISSR-nested PCR and selectively amplified microsatellite. 38 primers were designed by Primer 5.0. Polymorphism of these primers were shown on some varieties, include 'Xiaohuangli'. 28 primers showed polymorphism alleyes.2. A clustered dendrogram of 31 varieties of plum was obtained by mictosatellite marker and SSAP marker based on microsatellites. By mictosatellite marker, 4 European plum varieties were clustered to one subgroup,however,there were some varieties of different speciess in the subgroup.By SSAP marker, 4 European plum varieties were clustered to one subgroup without any varieties of different speciess. Thus we think that SSAP marker based on microsatellites was more effective than mictosatellite marker in clustering analysis.3. We put forward a new simple strategy based on the technique of sequence special amplify polymorphism(SSAP) for developing microsatellite markers. We used SSAP technique based on the complex repeat types microsatellite to find out the different fragments from different varieties or species groups. After that we cut them from polypropylene amine gel and transformed them to SCAR. While we sequenced them and designed special primers from the sequences.they were transformed to special microsatellite markers.With this method,we developed a microsatellite maker to differentiate the two varieties of 'Xiaohuangli' and 'Gaixiandali' plum.
