| The method of random sampling in typical population and whole-population sampling in central area were applied. Luxi yellow cattle(n=80), Bohai black cattle(n=50), Mountain buffalo(n=55) and Holstein cow(n=164) were sampled. The restriction polymorphisms of MHC-DRB3.2 in 4 populations of Bovinae were investigated by Semi-Nest PCR with enzyme of Haeâ…¢and BstYI. The results revealed as follow:1. There were extensive polymorphisms in the site of Haeâ…¢in 4 populations, but Mountain buffalo had fewest alleles in 4 populations. Extensive mutations also existed in the site of BstYI in Luxi cattle, Bohai cattle and Holstein cow, with homomorphism in Mountain buffalo.2. Statistical analysis showed that polymorphisms in the site of Haeâ…¢did not fit with Hardy-Weinberg equilibrium in Luxi yellow cattle and Mountain buffalo populations. Polymorphisms in the site of BstYI in Luxi yellow cattle, Bohai cattle and Mountain buffalo fitted with or almost reached Hardy-Weinberg equilibrium.3. Four populations are clustered by the methods of NJ and UPGMA based on DA and Roger distances respectively. The results are same to population state.4.This study indicated the PCR-RFLP system of cattle can be applied in the research of buffalo.5. This study also indicated that the decrease of population size may result the genetic disequilibria. The polymorphism of MHC-DRB3.2 in Mountain buffalo is less abundant than cattle. |
