Guangxi Bama Mini-pig, a highly inbred Mini-pig strain developed from Bama Xiang pig, was characterized by its miniature body size. No report had been observed previously on the molecular genetic mechanism of the body size in these animals. In the present study, growth hormone (pGH) and growth hormone receptor (pGHR) genes assumed as two candidate genes affecting the small body size of Bama Mini-pig were cloned and analyzed with the purpose to survey the genetic marker of miniature size of this animal strain. The primers were designed base on the pGH complete sequences and pGHR cDNA released at the GeneBank. The genome DNA extracted from Guangxi Bama Mini-pig ears tissue was used as template to amplify the full sequences of pGH gene. The pGHR extracellular and intracellular domains sequences were amplified by the RT-PCR used the total RNA as template purified from Guangxi Bama Mini-pig liver tissue. The amplified products were ligated into pMD 18-T vector after purification. The recombinants were verified through the method of PCR, restrictive cleavage and sequences survey, respectively. The results showed that the complete sequence of GH and the GHR...
