Abstract/Summary:
Polysaccharides is a kind of common biomacromolecules in organism body.It not only participates the composition of cytoskeleton ,but also is important component of many endogenous bioactive polymere.The research has found that the polysaccharide and its complexs participate in all kinds of cell vital movement.Besides anti-tumor,anti-virus ,anti-oxidizes, hypoglycemic, anti-lipemic,anti-radiation and so on,modulation of polysaccharide on immunologic function has been the hot topic in recent years.Potentilla anserine polysaccharide(PAP) is a nature polysaccharide extracted from potentilla anserine. This study summitted to the primary research of extracting and the pharmacological action of PAP.The Potentilla anserine polysaccharide was extracted with boiling water and precipitated by alcohol.Its content was determined by phenol-sulfurin acid method.Its chemical characteristics was verified by Molish reaction,iodine-potassium iodide reaction,ninhydrin reaction and infrared spectrum.Results were as folloaws:the extraction rate was 6.22%.The total content was 49.45%.The polysaccharide consists of four types of monosaccharides and hasα-configurations,and didn,t contain amylum and protein or amino acid.Immunosuppressive mouse model was established by cyclophosphamidum (100mg/kg).The animals were treated with PAP at different doses groups(50,100,200mg/kg) , the index of thymus and spleen,peripheral hemogram,organizational structure of thymus and spleen and ultrastructure of thymus were observed.The Results showed that PAP increased index of thymus and spleen of normal mouse as well as index of spleen of immunosuppressive mouse.The action of PAP on peripheral hemogram of immunosuppressive mouse was not sigificant.PAP improved and fighted apparently against damages induced by Cy according to observations in organizational structure and ultrastructure of thymus and spleen in immunosuppressive mouse.And this action presented certain dose-dependent relationship.To observe effects of the polysaccharide on reactive oxygen specie(ROS) ,dexamethasone was used as immunosuppressor to establish immunosuppressive mouse model. the index of thymus and spleen ,total supperoxide dismutase activiyty(T-SOD),content of nitrogen oxide(NO),the anti-superoxide anion(O2·-)...
