The thesis studied the polymorphism of BoLA-DRB3.2 alleles and analyzed the genetic correlation between BoLA-DRB3.2 alleles and economic traits of bovine (such as SCC, yield of milk, lipid percentage and protein percentage in milk)with static methods. It also inquired into the theoretical possibility of using BoLA-DRB3.2 alleles as genetic marker in marker assist selection at the resistance to mastitis.The polymorphism of BoLA-DRB3.2 alleles in some China Holstein populations was performed with semi-nest PCR-RFLP. After being digested with restriction enzymes Haeâ…¢, BstYI and Rsaâ… ,the products of semi-nest PCR-RFLP were segregated with 14%PAGE. Results indicated that semi-nest PCR could effectively and specially amplified the goal fragment. Six different restriction patterns as Haeâ…¢-a, Haeâ…¢-b, Haeâ…¢-d, Haeâ…¢-e, Haeâ…¢-f and Haeâ…¢-h were observed when the 284bp PCR products were digested with Haeâ…¢restriction enzyme. The corresponding frequencies of these patterns were 0.598,0.238,0.049,0.037,0.061 and 0.017,respectively.
