| 44 main cultivars of ramie were used to extract DNA by SDS,CTAB and Small sample extraction method.The results showed that CTAB was fast and simple for ramie's DNA extraction.80 random primers were used to amplify the total DNA from44 genotypes. The results showed that23 primers produced reproducible amplified products and the length of the products ranged 0.1~2.2 kb. Total 112 bands were identified. NTSYS software was used to analyze the similarity coefficients. 44 cultivars could be divided into 4 types , the genotypes of every type and group are familiar with each other in geographical distribution.SAS software was used to analyse 8shape and yield characteristics and4 quality characteristics of 44 varieties. The former characteristics were divided into 4 types by cluster anylasis, the genotypes are familiar with each other in geographical distribution.The later characteristics were divided into 4 types , and the genotypes are familiar in geographical distribution as well.All results were compared and showed that many similar outcomes could be obtained by using 3 different methods. 34 cultivars were similar with each other in molecular mark and quality characteristics, 33 cultivars in molecular mark and shape and yield characteristics. Additionally, these similar cultivars were also similar in geographical distribution.The results not only disclosed the genetic relationship of donated cultivars, but also affirmed that the experiment was reliable and accurate. |
