| Soybean flowers and pods abscission is not only the self-regulation of soybean growth and reproduction, but also an important restriction factor of yield. As one of hydrolases controlling organs abscission soybean abscission cellulase gene (GmAC) has an important role during soybean flowers and pod abscission. Expression of GmAC was regulated by the hormones and the unfavorable circumstances, so it has a significant academic meanings and applied values to carry out research on the mechanism of the hormones and the unfavorable regulating expression of GmAC, and their impacts on the flowers and pod abscission. Four soybean varieties ZaoShu18(ZSh18),KeXin3(KX3), YouChu4(YCh4)and Kefeng 1(KF1)were used in this research. Through the methods of RT-PCR,Northern blot and analysis and detection of several physiological and biochemical indexes, expression of GmAC in the flower and pod abscission zone cells,the abscission rate of flowers and pods and quantity of endogenous ethylene releasing of soybean different species under the conditions of three hormones(ethylene,ABA and IAA) and three unfavorable circumstances(high temperature,drought and salt) treatments, furthermore , expression of HSP70 gene of transformed plant were detected, the results show the following:1. Expressions of GmAC gene were respectively affected by the three hormones according to the different concentrate treatments. Whereas generally speaking, ethylene and ABA or both enforce expression of GmAC gene, but IAA restrains it. When the most favorable concentrate hormones treated soybean four species, expression of GmAC gene changes according to different soybean species. Increasing range of GmAC gene expression for low abscission rate species(ZSh18 and KF1) is more obvious than high abscission rate species(KX3,YCH4) when treated by ethylene or ABA, on the contrary, IAA affects more distinctly for high abscission rate species.2.
