Cryptosporidiosis is caused by Cryptosporidium spp which is a diarhoea disease in humans and domesticated animals. Cryptosporidium can infect hosts and cause disease lonely or companied with other pathogen, which was main by peroral infection. With the water contamination, the infection rates are climbing up in resent years. And infection of patients with immunodeficiency will be fatal. Cryptosporidiosis spreads out through Australia. U.S.A, Middle South America, Asia, Africa and Europe continents. Cryptosporidium exists world-widely. 1%-2% of patients with AIDS died from Cryptosporidium infection. The infection risk is up to 15%-+49% for those high risk-groups (i.e. infant, patients with immunosuppression and immunodeficiency). It's been reported that the rates of infection for developing and developed countries are 4-20% and 0.6-20% respectively. And for China, it's 1.4%-13.3%. The scope of host for Cryptosporidium is very wide. Presently, we have known Cryptosporidium can infect more than 170 different animals, including 79 mammals, from tropical to temperate zones. Separations from fish, amphibia and reptiles have been also reported.The characteristics of wide-scope of host and many species result in many different genotypes in conspecific animals which presents difficulty in epidemiologic survey. In late 90s, with the development of Molecular Biology, the characteristics of epidemics for Cryptosporidium at molecular level can be understood further. And so far, studies on molecular epidemiology of Cryptosporidium are practiced in different countries and regions, including many institutes world-wide. The studies focus on animals that closely relevant with mankind, i.e. swine, cattle and caprine.At present, gene sequences of 18SrRNA, acetyl coenzyme A synzyme, HSP70 and actin are used in genotyping and epidemiological analysis of Cryptosporidium.18SrRNA gene sequence is considered as a very effective marking molecule due to its high level of conservation in the evolutionary process. In recent years, with the help of 18SrRNA, many researchers from foreign countries practice studies on genotype, molecular epidemiology and phyletic evolution relationship of Cryptosporidium. Similar studies are also practiced in China.In this study, two pairs of primer for Nest-PCR were designed. And a specific fragment of Cryptosporidium 18SrRNA sequence was amplificated.
