Study On Antiviral Activity Of The New Preparations Of Hypericin On AIV In Vitro

Killing effect of the new preparations of hypericin on H9N2 subtype of low pathogenic avian influenza virus (LPAIV) was investigated by Klein-Defors test in vitro. 1,520 10-day-old SPF chicken embryos were divided into the new preparations of hypericin group(760 chicken embryos), Ribavirin control group(608 chicken embryos) and positive control group(152 chicken embryos). According to different consistency, the new preparations of hypericin group was divided into five subgroupings, and Ribavirin control group was divided into four subgroupings. To observe antiviral activity of the new preparations of hypericin in different time, every subgroupings was divided into two groups, one was inoculated after interacting for 10 minutes between the medicine and the virus, the other was inoculated after interacting for 30 minutes between them. The haemagglutinin titer was detected and the killing ratio was calculated. The results showed that the killing ratio of the new preparations of hypericin on H9N2 subtype of LPAIV was 0,0, 93.23%, 99.99%, 99.99 % when interacting not only for 30 minutes but also 10 minutes at the consistency from 0.0005mg/ml to 0.34mg/ml. It was concluded that the new preparations of hypericin had excellent inactivated effect to H9N2 subtype AIV, and the inactivated effect has none of acting time.To certify killing effect of the new preparations of hypericin on highly pathogenic avian influenza virus (HPAIV), killing test was investigated of the new preparations of hypericin on H5N1 subtype AIV in vitro, and the same test method, disposal of chicken embryos, while the number of chicken embryos was 1440. The results showed that the killing ratio was 0,0,94.37%,99.99%, 99.99% at the consistency from 0.0005mg/ml to 0.34mg/ml when interacting for 10 minutes, while interacting for 30 minutes, the killing ratio of it was 0, 32.39%, 95.42%, 99.99%,100% at the consistency from 0.0005mg/ml to 0.34mg/ml. The results also showed that the killing activity of the new preparations of hypericin on H5N1 subtype of HPAIV was associated with the consistency of the medicine and acting time.
