The purpose of this study is to detect the genetic variations of the goat FSHR gene. Forty-four boer goats, seventy-six Laiwu black goats, twenty-one boer×Laiwu black goats from Laiwu black goat breeding graduate school. we choose the FSHR gene as the candidate gene of the breeding trait, then amplified, sequenced, sequence blasted and PCR-SSCP tested the exon10 of different goat populations. The relationship of FSHR gene exon10 polymorphism and litter size was examined with General Linear Model(GLM) of SAS. The result were as follows:1. The exon10 of FSHR was amplified by PCR using genomic DNA of boer goats and Laiwu black goats as the template and designing primers according to the sequence of the cow receptor gene .The sequence was determined and analyzed according to cloning ,and the sequence was compared with that of cow FSHR gene . The results indicated that sequence length of FSHR gene was the same and was 1233bp among the three species . The exon10 sequence of the FSHR gene of the LaiWu black goat and boer goat compared with that of the cow, the homology were 97.49% and 97.41% respectively ,and each has 31 and 32 mutated bases and caused 10 amino acid code changes .While the exon10 sequence of FSHR of boer goat compared with LaiWu black goat,the homology was 99.60%,there were 3 mutations between them,that caused amino acid code changed respectively at +1184bp and +1365bp of the exon10 sequence of the FSHR.2. To detect DNA variation of goat FSHR gene, the sequence of exon10 has been studied by PCR-SSCP. Among the eight sub-sequences of this region, E10-1(198bp) was found to be polymorphic. It was showed three genotype (AA,BB,AB) in boer goats,four genotype (AA,BB,AB,DD) in Laiwu black goats and four genotype (AA,BB,AB,DD) in boer×Laiwu black goats. The genotype frequency of AA was the biggest among the three goat species. After sequencing the E10-1, G→T and A→G transitions were identified at the sites of 14527 and 14566.
