Techniques such as candidate genes, microsatellite marker linkage analysis become more and more important because they are simple, facility and applied.The aims of the present study that gene polymorphism associated with litter size and growth traits in Haimen , Huanghuai and second generation of Bore X Huanghua were: 1.to find the major gene for prolificacy in goat;2.to find microsatellite sites linking with prolificacy and provide theory bases for early selection in goat. Goats for experiments were from three populations, including 74 Haimen goat, 71 Huanghuai goat and 82 second generation of Boer goat x Huanghuai goat.1. Analyze goat prolificacy basing on prolificacy gene of sheepDNA samples were collected and amplified for bone morphogenetic protein receptor type IB gene (BMPR-IB), BMP 15 gene and GDF genes by PCR-RFLP method from 227 goat. The results showed that six markers weren't polymorphic in goat and none had mutation.;possibly the six markers weren't the major genes for prolificacy.2. Study on polymorphisms of microsatellites in three goat populationSix microsatellite markers named Lscv043, Bms2508, Gc101, 300U, Bulge5 and 471U which were closely linked to the FecB gene were analyzed in three goat population. Polymorphism information content (PIC), genetic heterozygosity (h), index of genetic diversity (H), and effective number of allele (E) were evaluated. Genetic distances were calculated between Three goat population.The results showed that microsatellite site Lscv043 and microsatellite site Bms2508 were high polymorphic; the genetic distance between Huanghaui goat and second generation of Boer goat x Huanghuai goat was further than that between Haimen goat and Huanghuai goat..3. Linkage analysis between six microsatellite markers and litter size, growthSix microsatellite markers which were closely linked to the Fec~B gene were analyzed in three goat population with their litter size or growth. Mean litter size at 2nd lambing for Lscv043 117/117bp were significanty higher than that for Lscv043 117/135bp,and total mean litter size for Lscv043 117/117bp. 107/117bp and 117/160bp were significanty higher than that for Lscv043 117/13 5bp in Haimen goat; mean litter size at 1st lambing and mean litter size at 2nd lambing for Ge101 200/238 were significanty higher that for Ge101 200/210 , 196/200(P<0.05),total mean litter size for Gc101 200/238 was the most significanty higher than total mean litter size for GclOl 200/210, 196/200(P<0.01);some body weights and sizes were significanty different for six microsatellite sites' genetype in second generation of Boer goat x Huanghuai goat.
