| Non-tuberculous mycobacteria is an environmental mycobacterial. It is mainly derived from the nature of the soil, water, dust, aerosols and animals. Most of the NTM are belonging to rot parasites. The environment in the world has a higher separation rate. At present, acquiring their infection from the environment is the main route of transmission. It has not been proven that people could spread. Non-tuberculous mycobacteria in biological traits vary with Mycobacterium tuberculosis. In morphology and staining characteristics with the bovine type of Mycobacterium tuberculosis, human type of Mycobacterium tuberculosis is very similar in pathogenicity and difficult to differentiate from Mycobacterium tuberculosis. But the non-tuberculous mycobacterial virulence is weak. Biochemical reactions are different and can be used to identify.Non-tuberculous mycobacterial disease and tuberculosis have similar clinical manifestations, such as systemic symptoms of poisoning and local damage. However, non-tuberculous mycobacterial disease symptoms are different for different organizations involved. Usually it can cause lung and pulmonary diseases, resulting in lymphadenitis, skin and soft tissue infections, bone and joint diseases, reproductive system, digestive system and nervous system lesions, and systemic disseminated disease. It can cause various animal and human infections. And especially cattle, pigs, the infection rate is higher. Cattle infected with this bacteria cause skin lesions, exudative mastitis, nodular mastitis and lymph nodes, especially the mastitis can affect milk production. After the pigs infected with certain pathogens, it can cause acid bacteria of pig disease and may spread to the liver, spleen and other substantive organs. It will affect livestock production and development and cause the domestic and international public health concern. In recent years, due to the gradual improvement of bacteriological analysis, immunocompromised inhibition of infection of the bacteria Non-tuberculous mycobacterial flora are changing, and the iatrogenic routes of infection are increasing. In clinical tests, the detection on frequency of non-tuberculosis mycobacteria increase, with the TB difficult to distinguish, However, there are treatment differences between the two. So to find the diagnosis-related peptide of Non-tuberculous mycobacterial disease both in epidemiology or in clinical diagnosis and treatment are of great significance..In this study, through the slaughter of cattle from the Jilin Province submandibular lymph nodes and mesenteric lymph nodes,12 strains of Non-Mycobacterium tuberculosis were isolated and cultured in the Middle brook 7H10 antigen medium and made into anti-serum, by Schaefer's agglutination and agglutinin absorption test for serotype identification, can be identified as 5 serotypes.Broken by ultrasonic method further extracted bacterial protein from five different serotypes of non-tuberculous mycobacterial, and then do the SDS-PAGE electrophoresis, after Coomassie brilliant blue R-250 staining, the number of bacterial peptides composition and molecular weight are analyzed. The results show that the cell protein electrophoresis profiles of the five have some similarities, bacterial peptide composition is consistent, and molecular weight is between 66 000~12 000.The bacterial protein of five serotypes contains four major protein bands of the same molecular weight, molecular weight were 65 000,45 000,38000,28000, the differences are mainly in the molecular weight which are 31000,25000,16000,12 000. And by Western blot in-depth study of its immunogenicity and the existence of cross-immunogenicity, it showed that in the bacterial polypeptide composition not only contains similar peptides zone, but the main antigenic peptides that have a similar immune response. This indicates that the major antigenic peptide has the similar structure. The molecular weight, which are 65000,45000 and 28000,is the more important cross-immunogenic proteins. This study is to develop more effective vaccines and to provide better references on disease prevention, prevent harm to human health and accelerate the development of animal husbandry in our province. |
