As the seventh member of Fibrolblast Growth Factors (FGFs) family, Keratinocyte Growth Factor (KGF) is a specific mitogen for epithelial cells, which can induce differentiation, proliferation and wound repair. It has a unique significance to clarify the details of upstream signaling pathway regulating KGF transcription. Firstly, we developed a novel chromosome walking method named as the Restriction-Site-Finder PCR, which derived from the SiteFinder PCR. By using the Alul site as an anchor site, we then isolated promoter of the red deer KGF gene for further analysis of cis-element. The result showed that the homology was high within major sites between promoter of human KGF and that of red deer KGF, but upstream of this region sequences diverged very much. Meanwhile we confirmed some important cis-elements like ERE, GRE, AP-1, which can prove important information to analysis the KGF signaling pathway.
