Prevention and control of rabies mostly delied on immunization, and rabies vaccines are commonly used as live attenuated vaccine, the attenuated live vaccine of rabies virus is generally Flury-lep strain. Therefore it is necessary to comprehensive evaluated the ecological characteristics of rabies virus Flury-lep strain to ensure and improve the quality of rabies vaccine.In this study, we observed rabies virus Flury-lep strain which shaped of a bullet-shaped particles or cone In BHK-21 cells, chick embryo, and cultures of mouse brain by scanning electron microscopy. Under the electron microscope, virus particles were blunt or sharp tip of the arc, and flush the bottom of the virus particles. Therefore, roughly the same shape, are bullet-shaped or cone, but there are some small differences, electron microscope images showed in:BHK-21 cell and chick embryo which shape of the bullet, and cultured mouse brain, there are many virus-like particle shape of the bullet.BHK-21 cell culture and embryo culture rabies virus Flury-lep strain which the virus particle shape and size are more consistent, and cultured mouse brain rabies virus Flury-lep strain which the virus particle shape and size were quite different. Virus particles of different cultures in the size difference is obvious. From the rabies virus Flury-lep strain in different cultures, the virus particle morphology is more stable. The rabies virus Flury-lep strain vaccine bestly would be to use the proliferation of BHK-21 cell cultures.Secondly, the study also examined the rabies virus strain Flury-lep hemagglutination of goose erythrocytes of which cultured in BHK-21 cells and chicken embryo. At 4℃, HA test results showed that:Flury-lep strain could agglutinate goose red blood cells, BHK-21 cell cultures of HA titer were regular and higher, four samples of the HA titer:1:256,1:256,1:256,1:256; chick embryo cultures of HA titer dependent objects were lower priced regular and valence, four samples of the HA titer:1:16,1:8,1:8,1:8. From the results, there are significant differences in BHK-21 cells with rabies virus Flury-lep strain better than with the proliferation of chick embryo rabies virus Flury-lep strain. Therefore, the preparation of rabies virus strain Flury-lep attenuated vaccine, betterly use the BHK-21 cell strain of rabies virus Flury-lep.Finally, the rabies virus Flury-lep strain glycoprotein gene was cloned and sequenced and compaired with other rabies strains glycoprotein gene and amino acid sequence. According to the sequencing of the rabies virus Flury-lep strain glycoprotein gene nucleotide sequence, drived Flury-lep strain glycoprotein amino acid sequence using DNAstar software. And compaired with other 10 rabies strains glycoprotein gene and amino acid sequence. The nucleotide sequence homology was 90.2%~97.5 % amino acid homology was 90.4%~95.4%.
