Rabies is a fulminating infectious disease and is very harmful to human beings and animals. Vaccination is the only effective current prevention and treatment of rabies. However, as a result of its short incubation period, the current tissue culture vaccine can not provide 100% protection. And rabies virus glycoprotein plays a very important role in the prevention and treatment of rabies, and is the only antigen to induce organism to produce neutralizing antibody. IL? is an important immune function regulatory molecule, and can provocate the proliferation and activation of B and T cells and strengthen the immune response, and it can also act as molecular adjuvant. According to the properties of rabies virus glycoprotein and IL?, the specific glycoprotein gene was cloned from RNA of rabies virus gene group in this experiment. After the sequence check and analysis of glycoprotein gene, we found its whole length was 1575 bp and there were 524 encoded amino acids and four mutants in the whole GP gene. Their sites were , the 55th 370th ? 566th ?, and 596th ?. And these points were respectively mutated into A, G ,T and A, and the first three mutations, due to the degeneracy of nucleotide, did not affect its encoding of amino acid. While, after the mutation from ? into ?at the 596th point, the codon TGG was mutated into TGA as the termination code , which affected the translation and expression of the nucleotide sequence. By the fixedoint mutation of ?into ?at 596th the integerity of glycoprotein antigen might not be affected. By cloning glycoprotein gene into the multilone points of pCI-neo expression vector, the recombination plasmid of pCI-neorabG was established. And the recombination plasmid of pCI-neoIL2 was successfully established by the same way. Vaccinated with these two recombiriation plasmids in mice, we found rabies virus antibody could be determined in four mice in pCI-neorabG group, while with the combination of pCI-neoIL2, 6 mice of 10 produced rabies virus antibody, the antibody titers of both groups are low, while the controll group is negative. The lymphocyte proliferation experiment results show that with pCI-neoIL2, the number and appearance of lymphocyte become more and larger. These results show pCI-neoIL2 can play a part in rabies DNA vaccine as a molecular adjuvant.
