Take the Oula Tibetan sheep, Ganjia Tibetan sheep, Qiaoke Tibetan sheep, Poll Dorset, Genmeny marino and Small Tailed Han sheep in six sheep group of 404 individuals as the research object, detected the genetic polymorphism of the CLPGSNP loucs of Callipyge gene and partical sequence of CAPN1 gene by using PCR-SSCP, analysised the correction of growth traits (weight, length, body height, chest circumference and chest depth) and polymorphic loci by least square method, provides basis theory for molecular genetics of sheep breeding. Results are as follows:1. The results of PCR-SSCP about P primers of Callipyge gene indicted that: there are AA, AB and BB three genetypes in the six sheep group, AA genetype is the advantage genotype and A alleles is the advantage alleles.The results DNA sequencing showed:there is a novel C to T mutation in 176bp.2. The results of PCR-SSCP about L1 primers of CAPN1 gene indicted that: there are AA, AB and BB three genetypes in the six sheep group, A alleles is the advantage alleles of Poll Dorset, B alleles is the advantage alleles of the remain sheep. The results DNA sequencing showed:there is a novel A to G mutation in 190bp in the fifth introns of the L1 amplified fragment; there is a novel A to G mutation in 260bp in the sixth introns, there is no amino acids changed so this is a synonymous mutations.3. The results of PCR-SSCP about L2 primers of CAPN1 gene indicted that: there are AA, BB, CC, AB, AC and BC sixgenetypes in the six sheep group, but only detected CC genetype in Oula Tibetan sheep. A alleles is the advantage alleles The results DNA sequencing showed:there is a novel A to G mutation in 21bp, T to C mutation in 24bp, G to A mutation in 42bp, G to A mutation in 122bp, C to T mutation in 146bp, the later two'mutation in exon9, but there is no amino acids changed so this is a synonymous mutations.4. Genetic analysis indicted that:the heterozygosity is lower in the detection Callipyge genes polymorphic loci of six sheep group,so the polymorphism is low, tests of Hardy-Weinberg equilibrium confirmed that all sheep group was in equilibrium(P>0.05) expect Small Tailed Han sheep. The heterozygosity is hinger in the detection CAPN1-L1 and CAPN1-L2, show moderate polymorphism.Tests of Hardy-Weinberg equilibrium confirmed that all sheep group was in equilibrium (P>0.05) expect Poll Dorset.5. Analysised the correction of growth traits and polymorphic loci by least square method, the results showed that:the polymorphic loci of Callipyge gene's has no significant with growth traits.The AB genetype's chest deep significantly higher than BB's of Oula Tibetan sheep of CAPN1-L1 (P<0.05), The BB genetype's chest deep significantly higher than AB's of Poll Dorset of CAPN1-L1 (P<0.05), the polymorphic loci of CAPN1-L2 has no significant with growth traits.
