Objective:To evaluate effect of sevoflurane on the expression of inflammation cytokine in neuroglioma cells of cultured mice.Methods:Microglia cells abtained from newborn(2-3 days) C57BL/6 mice were se parated and randomly divided into 4 groups(n=10):control group(Control);sevoflura ne group(Sevoflurane); NF-ΚB inhibitors group(PDTC); NF-ΚB inhibitors group+sevoflurane group(PDTC+Sevoflurane). Oxygen(21%), Carbon dioxide(5%) and s evoflurane(4.1%) were delivered from an Drager anesthesia machine to the microgl ia cells sealed in a plastic box of the two groups. The two groups are Sevofluran e group and PDTC+Sevoflurane group in which the later group had been given P DTC 1 hour before sevoflurane treatment. A Drag gas analyzer was used to conti nuously monitor the concentrations of the O2, CO2 and sevoflurane respectively. Ib a- 1 immunofluorescence staining method was used to the purity identification of microglia in mice. We examined the expression of IL-6 and TNF-α(by Western Blotting Analysis and ELISA)and activation of NF-ΚB(by Western Blotting Analy sis) at 2,4 and 6h after sevoflurane treatment.Results:Sevoflurane increases levels of IL-6 and TNF-α through activates NF-ΚB signaling pathway.Conclusion:These studies suggest that NF-ΚB signaling pathway could contribute to the increasing expression of inflammation cytokine.
