| Objective: To study the association between the D543 N polymorphism of NRAMP1 gene,-292 and +1320 polymorphisms of CISH gene and susceptibility to tuberculosis in Tibetan population. Methods: In the case- control study, 135 cases of Tibetan active pulmonary tuberculosis patients and 143 healthy Tibetan controls were recruited. PCR-RFLP method was applied to amplify gene and discriminate the genotype. SPSS19.0 statitical software was used to analyse the data. Grouping by the genotypes, to compare the percentage chi-square test was apllied. The OR and 95 CI were also calculated. Results: 1. With repect to the D543 N locus of NRAMP1 gene, in the case group there were 108 cases(80%), 26 cases(19%), 1 case with G / G genotype, G / A genotype and A / A genotype respectively. Meanwhile, in the control, there were 130 people(90%), 12 people(8.4%) and 1 people(0.7%) with G / G genotype, G / A genotype and A / A genotype seperately. Comparing to G/G genotype, distribution of G / A genotype in case and control group was significantly different(P<0.05)(OR =4.408,95%CI,2.137-9.093). 2 Regarding-292 locus of CISH gene, in the case group, the number of patients with A / A genotype, T / A genotype and T / T genotype were 58(43%), 65(48.1%) and 12(8.9%) respectively. At the same time, in the control group, the number of people with A / A genotype, T / A genotype and T / T genotypes were 66(46.1%), 62(43.4%) and 15(10.5%) separately. In terms of +1320 locus of CISH gene, in the case group, the number of patient with A / A genotype, C / A genotype and C / C genotype were 81(60%), 51(37.8%) and 3(2.2%) respectively. And in the control group, there were 80 people(56%), 56 people(39%), 7 people(5%) with genotype of A / A, C / A and C / C respectively. Comparing to A/A genotype, the distribution of C / A and C / C genotype in case and control group were not significantly different. Conclusions:(1) The D543 N polymorphism of NRAMP1 gene was associated with susceptibility to tuberculosis in Tibetans population. Further more, G / A genotype may be tuberculosis susceptibility genotype in Tibetan population.(2) There was not association between-292 and +1320 polymorphisms of CISH gene and susceptibility to tuberculosis in Tibetans population. |
