Objective:To observe the influence and mechanisms of Genistein on mouse model of airway inflammation in asthmatic mice. Methods:24 SPF level BALB/c female mice were randomly divided into control group,asthmatic group,methylprednisolone group and genistein group.Each group contained six mice.Mice of asthmatic group,methylprednisolone group and genistein group were injected by 0.1ml sensitizer(300μg OVA) in section 0、7、14 day.On days 15,we began intraperitoneal injection of drugs to section 23day:mice of methylprednisolone group was injected by 0.1ml methylprednisolone(800μg methylprednisolone),genistein group was given intraperitoneal injection of 0.1ml genistein(genistein 20mg/kg),asthmatic group was given 0.1ml saline injection.In the last three days,one hour after administration,mice were given 10ml 1%OVA aerosol inhalation(30 minutes) once a day.The control group was injected by saline in section 0、7、14 day,and from section 15 day,we began intraperitoneal injection of saline to section 23 day. In the last three days of administration,one hour after administration,mice were given 10ml saline aerosol inhalation(30 minutes) once a day.24h after the last challenge,we removed eyes of mice to collect blood and then collected bronchoalveolar lavage fluid (BALF). The level of interleukin(IL)-4、IL-13 in BALF,total immunoglobulin E(IgE) levels in serum and lung tissue thymic stromal erythropoietin(TSLP)were measured by enzyme-linked immunosorbent assay(ELISA).Finally,we used method of hematoxylin - eosin (HE) to assess cell infiltration inflammatory of non-lavage lung tissue.Results:1.The levels of IL-4,IL-13 in BALF and TSLP、total IgE in serum were increased in asthmatic group compared with in the control group (P<0.05).2.The levels of IL-4、IL-13 in BALF and TSLP、total IgE in serum of methylprednisolone group had a decrease than that in asthmatic group(P<0.05).3.The levels of IL-4、IL-13 in BALF and TSLP、total IgE in serum were decreased in genistein group compared with the asthmatic group(P<0.05).4.There were no significant differences on the levels of IL-4、 IL-13 in BALF and TSLP、total IgE in serum between genistein group and methylprednisolone group(P>0.05).5.Lung tissue of control group of mice had no inflammatory cell infiltration.Lung tissue of asthmatic group of mice had great amount inflammatory cell infiltration and airway spasmed obviously. Lung tissue of methylprednisolone group and genistein group of mice had few inflammatory cell infiltration and airway spasmed slightly.Conclusion:Genistein could inhibit Th2 advantage immune mediated by thymic stromal lymphopoietin (TSLP) in asthmatic mice.
