Objective: The experimental research chose the model of rats which had been provoked myocardial ischemia by isopropylarterenol hydrochloride with Western Blot testing technique. We observed the function selecting acupoints along the meridian in regulating and controlling the Protein Kv1.4、Protein Kv4.2、Protein Kv4.3 on the transient outward potassium channel and auxilin KCHIP2 on the Ito channel. Through the study of the possible mechanism, which the condition of myocardial ischemia was improved by acupuncture, we tested the specificity of acupoints function along the meridians, and provided the scientific basis for the treatment of myocardial ischemia with Electroacupuncture. Methods: The 72 rats, which were SPF level SD, healthy and male, randomly selected 10 rats as comparable group. Besides the comparable group, the myocardial ischemia models were built in the rest 4 groups with subcutaneous multi point position injection of isopropylarterenol, while the comparable were injected the equal amount of physiological saline in the same position, the modeling of the 22 rats were removed unsuccessful. The other 40 rats were distributed to 4 groups according to randomly numerical table. There were model group, LIEQUE group, NEIGUAN group, non-acupoints group, and 10 rats in each group. The rats in LIEQUE group,NEIGUAN group, non-acupoints group were needled obliquely along the meridians according to the acupoints of each group with‘Hua Tuo’ brand Number 36 needle(0.20mm×25mm), which was 2mm subcutaneous prick, connected 6805-D electric acupuncture apparatus, dilatational wave operated, needle retained for 20 minutes, each time a day, 7 times in a row.The electrocardiogram of rats in each group was inspected by BL-420 s biologically functional experiment system. The expressive transformation of the relative Protein Kv1.4、Protein Kv4.2、Protein Kv4.3 in myocardial cells was inspected by western blot which in the transient outward potassium channel. Results: The transformative tendency of four protein’s index appearedidentical. Both the protein expression of NEIGUAN group and LIEQUE group rose(P<0.05), while non-acupoints group and model group showed some disparity but had no statistic meaning. Meanwhile the protein expression of non-acupoints was lower than NEIGUAN group and LIEQUE group, but the comparation between NEIGUAN group and LIEQUE group had statistic meaning(P<0.05),the protein expression of NEIGUAN group was higher than LIEQUE group, lower than the LIEQUE group.Conclusion:1.Electroacupuncture of NEIGUAN acupoints and LIEQUE acupoints have the effectiveness on restricting the rats electrocardiogram transformation which was caught myocardial ischemia by isopropylarterenol hydrochloride.2.Both the electroacupuncture of NEIGUAN and LIEQUE can increase the relative Protein Kv1.4、Protein Kv4.2、Protein Kv4.3 of the transient outward potassium channel belonging to rats myocardial cells, moreover, the NEIGUAN is more effective than LIEQUE.3.The electroacupunture of NEIGUAN has the specificity to treat myocardial ischemia along the meridians.
