Objective: To establish the animal model of spleen deficiency and dampness stagnation type of Functional Diarrhea(FDi) and the study of its pathogenesis,and to discuss the mechanism of Shenlingbaizhu powder in treating the rats of spleen deficiency and dampness stagnation type of Functional Diarrhea.Materials and methods: To select 90 male Wistar rats at the age of 4 weeks,making model by eating and over exercising.Make them standing on small flat for 10 hours every night,and feed them with high lactose for 2 weeks.After the success of modeling,divided into blank control group(A),model group(B),positive drug(Dicetel)control group(C),high dose of Chinese medicine group(D), middle dose of Chinese medicine group(E),low dose of Chinese medicine group(F),15 rats in each group.The rats were given Shenlingbaizhu powder by oral gavage for 2 weeks,and they were killed and taken after treatment.To observe the change of behavior of the rats,histological examination, calculate small intestinal advancing rate,check content of 5-HT,MOT,SS of the serum,and detect the expression of AQP-3,AQP-8 in intestinal tissue.Results: 1.Compared with normal rats,rats of spleen deficiency and dampness stagnation type of FDi had loose stools,weight loss,less activities,slow increase of weight(P<0.05),food intake decreased significantly. 2.Compared with blank control group,except rats of low dose group was not obvious in the recovery,rats of other Chinese medicine group and positive drug control group recovered in all indexes, and there is no significant difference between high dose of Chinese medicine group and positive drug control group. 3.D- xylose in serum of rats in model group was significantly lower than that of rats in blank control group(P<0.05),and Shenlingbaizhu powder in high dosageand Dicetel could increase D-xylose in serum of model rats;small intestinal advancing rate of rats in model groups was significantly higher than that of blank control group(P<0.05),Shenlingbaizhu powder in high dosage and Dicetel could reduce small intestinal acvancing rate of model rats. 4.5-HT in serum of rats in model group was significantly higher than that of blank control group(P<0.05),Shenlingbaizhu powder in high dosage and Dicetel could reduce 5-HT in serum of model rats significantly(P<0.05); MOT in serum of rats in model group was significantly lower than that of rats in blank control group(P<0.01),Shenlingbaizhu powder in high dosage and Dicetel could increase MOT in serum of model rats(P<0.01);SS in serum of rats in model group was significantly higher than that of blank control group. 5.The expression of AQP-3 and AQP-8 in colonic mucosa of rats in model group was significantly lower than that of rats in blank control group; Shenlingbaizhu powder in high dosage and Dicetel could increase the expression of AQP-3 and AQP-8 in colonic mucosa of model rats.Conclusion: 1.Model of spleen deficiency and dampness stagnation type of Functional Diarrhea(FDi)could be made by method of high lactose diet and standing on small plate. 2.Shenlingbaizhu powder has significant therapeutic effect on functional diarrhea of spleen deficiency and dampness stagnation type,and the effect of Shenlingbaizhu powder in high dosage is superior.
