Purpose:ThroughacupuncturemyocardialischemiamicemodelonNeiguan,Liequeand Non-acupoint to observe the changes of cardiac muscular tissue CaM and CaMKⅡ and L-type calcium channel alpha1on gene expression levels,to revealacupuncture Neiguan forming the protection mechanism of myocardial ischemia atthe genes level,providing experimental basis that electric acupuncture onNeiguan have good adjustment function to myocardial ischemia.Material and method:40healthy adult ACCN3knockout mice according torandom number table method randomly divided into blank control group,model group,Neiguan group,Lieque group,Non-acupoint group,each group has eight.In additionto the blank control group,other groups using multiple point injection isopropyl adrenaline hydrochloride injection(85mg/Kg)under the skin,myocardialischemia mice model is set up; Blank control group all inject the saline injectionin the same place.After the success of building model,taking Neiguan,Lieque,Non-acupoint separately,after routine disinfection,using Huatuosterile brand0.18x25mm to acupuncture,Neiguan,Non-acupoint using the needlebody and skin is90°Angle of stab method,Lieque with suitable Angle of30°oblique thrust method,2-3mm,to Pierce the skin connection electric acupuncture,cupping treatment to choose density wave,2/50HZ frequency,intensity is2-4ma,taking mice body quivering as a standard,retaining needle for20minutes,1timea day,acupuncture for seven consecutive days.Record the type leadelectrocardiogram Ⅱ T wave amplitude changes.Acupuncture groups in seven daysto be put mice to death,the application of real-time fluorescent quantitativepolymerase reaction technology(Real-time PCR)detection of myocardial tissuecalmodulin(CaM),calmodulin kinase(CaMK Ⅱ)and L-type calcium channel alpha1gene expression level. Results:1. Compared with blank control group,model group of myocardial tissue in miceCaM,and CaMK Ⅱ and L-type calcium channel alpha1of gene expression weresignificantly increased (P<0.01), with statistical significance.2. Compared with model group,the myocardial tissue of Neiguan group,Lieque group,Non-acupoint group mice in CaM and CaMK Ⅱ and L-type calcium channel alpha1gene expression of significantly reduced (P<0.01), with statisticalsignificance.3.Compared with Non-acupoint group,the Neiguan group has significant difference(p<0.01);contrast with Lieque group,the Neiguan group has statisticalsignificance (p<0.05).Conclusion:1.Electric acupuncture on Neiguan can significantly reduce the gene expressionof ischemic myocardial tissue by the closed cavity in CaM and CaMK Ⅱ and L-type calcium channel alpha1.2.The mechanism of treating myocardial ischemia using electric acupuncture onNeiguan,may be through the regulatory role of L-type calcium channel,reduce the"calcium overload".
