Purpose:To investigate influence of CYK formula for UC on CD4+CD25+Treg、TGF-β1level in colon of ulcerative colitis model in rats。To investigateinfluence of CYK formula for UC on P-pg level in colon of ulcerative colitismodel in rats. Compare the curative effect with SASP. To clarify the treatmentof ulcerative colitis and possible mechanism of action, in order to providetheory for the treatment of ulcerative colitis and practice basis.Material and method:Using the immune method to improving the UC rat mademould. Specific way of buliding the method is (TNBS) method. After the moldeing48hours,random kill a rat in each of the blank control group and the modelgroup. Observing the modling result.Taking the lesion site colon tissue forHE staining. Microscopic observation of the tissue,make sure that the modlingis successful and then continue the experiment. Confirm modling was success after2days then start to treatment. The treatment group was given the dose of10.8g/200g of CYK suspension, SASP group was given the injectable suspension of SASPwith the does of0.823g/200g, model control group and blank control group togive equal saline lavage. Onch a day, the treatmenta was totally of2weeks.Two weeks after the treatment of traditional Chinese medicine (CYK) enteritis,application HE dyeing observation the colon histologic changes; Flow cytometryinstrument detection UC rat peripheral blood CD4+CD25+Treg proportion;Immunohistochemical detection of colonic mucosa tissue expression of TGF-β1;Immunohistochemical method to detect the P-pg expressed in rat colon tissuechanges. Using SPSS18.0statistical software, all the data to mean±standarddeviation (x±s) show that conform to the normal distribution of data after ftest, compares the single factor analysis of variance between groups, do notaccord with normal distribution data using nonparametric test (rank and inspection) difference was statistically significant (P <0.01).Results:The experiment showed that after2weeks treatment, the treatment group ratshave no mucus excrement, and purulent blood.It’s general condition improvedmarkedly in the model group. Histopathological prompt treatment group rats coloncolon tissue inflammatory cells compared with model group significantly reduced,without glands is missing, no congestion, edema. Histologic grading treatmentgroup significantly lower than the model control group (P <0.01), the differencewas statistically significant. Prove that the traditional Chinese medicineenteritis CYK granules in treatment of UC effectively, and the curativeeffect.the proportino of CD4+CD25+Treg/CD4+T in treatment group rising higherthan model group in the peripheral blood (P <0.01), the difference wasstatistically significant, enteritis, can obviously increase the UC model inrat peripheral blood the proportino of CD4+CD25+Treg/CD4+T. Treatment groupTGF–β1expression in rat colon tissue compared with model control groupsignificantly decreased (P <0.01), the difference is statistically significant.P glycoprotein expressed in the treatment group rats colon tissue is blank wayincreased significantly (P <0.01), the difference is statistically significant.Compared with model control group decreased, but no significant difference, thedegree of TGF–β1is the condition of UC detection index, can reflect the UCactivity. Traditional Chinese medicine (TCM) enteritis, particles can inhibitthe UC model P-pg expression in rat colon tissue.Conclusion:1. The traditional Chinese medicine (TCM) enteritis kang granules therapy forthe treatment of UC is effective. Compared with western medicine SASP, enteritis,can be raised in the peripheral blood CD4+CD25+Treg/proportion of CD4+T, andby inhibiting the secretion of CD4+CD25+Treg, TGF–β1, regulate the body’simmune response, promote the Th1/Th2cell activation, adjust the balance ofthe anti-inflammatory and proinflammatory cytokines, restore the immune balance. This may be one of the mechanism of traditional Chinese medicine (TCM) enteritiskang Chinese medicine in the treatment of UC. UC pathogenesis process, CD4+CD25+Treg activation causes the TGF–β1secretion to increase, involved in theprocess of immune response, inhibit inflammation, TGF–β1is the condition ofUC degree detection index, can reflect the UC activity. Traditional Chinesemedicine (TCM) enteritis, particles can inhibit the UC model P-pg expressionin rat colon tissue.2. Traditional Chinese medicine (TCM) enteritis, can effectively reduce thelevel of P-pg in the colonic mucosa tissue of rats, delay the drug eliminationof bowel mucosa, enhance drug curative effect, reduce the resistance. CompoundChinese medicine for the treatment of UC in clinical are getting more and morewidely used, the experiment of traditional Chinese medicine (TCM) in regulatingcytokine balance, promote the immune balance only has carried on the preliminarydiscussion on the recovery mechanism, the pathogenesis and treatment of UC andthe influence of traditional Chinese medicine for its still needs more in-depthresearch and discussion.
