Effect Of Qianlongtong Capsule On Expression Of Bcl-2 And Bax In Prostatic Tissue Of Rats With Hyperplasia

[Objective]Exploration of action mechanism and influence of protein Bcl-2and protein Bax of BPH rat prostate by using Qian long tong capsule.[Method]1. According to the principle of the randomized,60rats were randomly grouped into blank control group, model group, Long bi shu group, Qian long tong low dose group, Qian long tong middle dose group and Qian long tong high dose group.2. Using the method of water substitution and electron balance to measure prostate volume,wet weight and index.3. Using HE staining of prostate tissues of rats microstructure changes and calculate the glandular epithelium and stromal area.4. Observe the effect of prostate tissue of rats Bcl-2mRNA, BaxmRNA expression by in-situ hybridization.5. Observe the effects of prostate tissue of rats Bcl-2, Bax expression by immunohistochemistry.[Results]1.The weight and the prostate wet weight of every rats group have statistically significant difference (P<0.05) before intervention.2.Each dose group of the rat prostate wet weight and volume in Qian long tong capsule group, comparing with model group, have statistically significantly different (P<0.05).And the Qian Long tong capsule high-dose group has previously differences in maximum, its wet weight and volume of the prostate were more close to normal group.3.Bcl-2expression of BPH rats are significantly higher than normal group, and the expression of Bax are significantly lower than normal group.After using Qian long tong capsule, they can be adjusted to normal levels. 4.Qian long tong capsule can reduce the Bcl-2mRNA positive cells expression and increase BaxmRNA positive expression in prostate tissue.[Conclusions]Qian Long tong capsule can effectively treat BPH rats, and its mechanism may be through inhibiting the Bcl-2expression and increase the expression of Bax, thereby promoting apoptosis of prostate.
