Effects Of Yangxian â…  On Hemorheology And Endometrial VEGF In Rats With Renal Deficiency And Blood Stasis Syndrome

[Objective]Based on the the TCM theory "kidney deficiency and blood stasis syndrome is the common pathogenesis of infertility" and copying the rat models with kidney and blood stasis syndrome,this study observes that assisted reproduction I prescription improves the general condition,the blood rheology and VEGF (vascular endothelial growth factor) in endometrial tissue, which explores the mechanism of assisted reproduction I treating infertility and provides the basis of the relevant laboratory in the clinical application, as well as establishes a theoretical foundation for further research.[Method]According to the physiological activity,50female rats, in proestrus stage, are randomly assigned to the blank control group, model group, positive control group, high-dose group and low dose group. Each group has ten rats. Blank group (normal saline group), model group (Hydroxyurea+saline), positive drug group (Hydroxyurea+compound danshen tablet), low dose group (Hydroxyurea+assisted reproduction I prescription) and high dose group (Hydroxyurea+assisted reproduction I prescription). Blank group rats are given5ml/kg saline lavage every day, the rest of the each group lavage by hydroxyurea450mg/(kg·d) once per day,7days in a row. At the end of the experiment, dosing the10%polymer dextran5ml/kg from the intravenous back quickly, by which the kidney and blood stasis model is established. Finally we gain the blood through the orbital venous plexus and detect hemorheology indexes, and quickly carotid off the rats, take out the uterus tissue. Then the number of spiral arteries in endometrial tissue is observed and pathology change of VEGF under a microscope.[Results]1. The influence of general situation of model rats with kidney deficiency and blood stasis syndrome, ponderal growth and index of uterus. The influence on the general situation of rats after treated by assisted reproduction I prescription:compared with the blank group, the spirit, activity, stamina, coat off, defecate, the respect such as tongue of model group rats are generally poor. Ponderal growth and index of uterus has significantly increase (P<0.05). Compared with model group, the general situation of rats in compound salvia tablet group, high and low dose group were improved to some extent.2. The influence of hemorheology of model rats with kidney deficiency and blood stasis syndromeThe influence on the hemorheology of rats:compared with the blank group, the hemorheology model group rats has the significant differences (P<0.05) or extremely significant differences(P<0.01).Compared with model group, all values of compound salvia tablet, high and low dose group were significantly differences (P<0.05) or extremely significant differences(P<0.01).3. The endometrial VEGF expression of model rats with kidney deficiency and blood stasis syndromeThe influence on VEGF area, total integral optical density and the mean average blackness expression of endometrial tissues:compared with the blank group, the VEGF area, total integral optical density and average blackness in model group are significantly reduced and extremely significant differences (P<0.01).Compared with model group, the VEGF area, total integral optical density and average blackness in compound salvia tablet,low and high dose group are significantly higher average expression and has significant differences (P<0.05) or extremely significant difference(P<0.01).[Conclusion]1. This research preliminary designs and builds model rats with kidney and blood stasis syndrome caused by the hydroxyurea lavage and caudal vein muscle injection.2. Different doses of assisted reproduction I prescription functioning as tonifying kidney and promoting blood stasis can improve normal condition of rats, increase the weight and uterine index.3. Different doses of assisted reproduction I prescription functioning as tonifying kidney and promoting blood stasis can obviously improve blood rheology of thick, sticky, coagulation, poly condition of the model rats with kidney and blood stasis syndrome.4. Different doses of assisted reproduction I prescription functioning as tonifying kidney and promoting blood stasis can promote the expression of VEGF in the endometrial tissue and the growth of spiral arteries.In a word, all the conclusions prompt that assisted reproduction I prescription has the function of tonifying kidney essence and promoting blood circulation to remove blood stasis, obviously improves the blood supply of uterine artery, promotes endometrium mature, which benefits the implantation of fertilized ovum.
