Objective:To establish model rats of Generalized Anxiety Disorder(GAD) by giving them predatory stress and Vogel’s conflict test,treat them using Danzhi Xiaoyao powder(DXP) with the effect of soothing liver, clearing heat and tonifying spleen and to discuss the pathogenesis of GAD and the intervening effect of DXP from the aspects of ethology and cerebral metabolism. To complete the theory about treating GAD with therapy of soothing liver, clearing heat and tonifying spleen further more.Methods:32 male Wistar rats were randomly allocated to four groups as follows:normal(distilled water),control(distilled water),Chinese medicine(DanzhiXiaoyao powder),western medicine(buspirone hydrochloride)(n=8 each). All groups except the normal were given predatory stress and Vogel’s conflict test which lasted for 21 days to establish GAD models, and from the 15th day, all groups began to be administrated by gastrogavage lasting 7 days. After modeling,We used EPM(Elevated Plus Maze)to evaluate all rats’behavior and their ratios of N-acetyl aspartate(NAA)/creatine(Cr), choline(Cho)/Cr and glutamate(Glu)/ Cr of bilateral prefrontal cortices and hippocampi were measured by highfield strong superconductivity(70T) animal MRI.Results1EPMCompared with the normal group, the OE% of the model group decreased apparently(P< 0.01). Both of the western medicine group’s OE% and the Chinese medicine group’s OE% increased obviously(P<0.01)compared to the model group. Each OT% of the other three groups showed decreased apparently when compared to the normal group(P< 0.01).The OT% of the western medicine group and the OT% of the Chinese medicine group were higher obviously than that of the model group (P<0.01).2The ratios of the metabolites in rats’bilateral prefrontal cortices and hippocampiCompared with the normal group, the model group’s NAA/Cr of bilateral prefrontal cortices decreased, while its Cho/Cr and Glu/Cr of the right lateral prefrontal cortex showed higher,those differences above were statistically significant(P<0.01).The Cho/Cr and Glu/Cr of the left prefrontal cortex had no obvious differences between the normal group and the model group. Compared to the model group,the Chinese medicine group’s NAA/Cr、Cho/Cr and Glu/Cr of the right lateral prefrontal cortex showed differences,with NAA/Cr increased and Cho/Cr and Glu/Cr decreased in specific (P<0.01; P<0.05),whereas there was no difference of those metabolites above in the left prefrontal cortex.Compared with the normal group about the ratios of metabolites in the right lateral hippocampus, the model group showed NAA/Cr and Cho/Cr decreased,Glu/Cr increased, which has statistical significance (P< 0.01);As for the ratios of the metabolites above in left lateral hippocampus, there had no difference between the normal group and the model group. As the ratios of metabolites in the hippocampi between the Chinese group and the model group,there had statistically significant differences in the right lateral hippocampus,with NAA/Cr and Cho/Cr increased and Glu/Cr decreased in specific, while there was no difference for the left lateral hippocampus.Conclusions1The exploratory and activity of the model rats established by the predatory stress and Vogel’s conflict test appeared declined, which indicated that the model rats became anxious and proved that the model was successful.2The results about the ratios of the metabolites in the rats’ bilateral prefrontal cortices and hippocampi tested by the technology of 1H-MRS showed that there existed differences especially in the right lateral between the normal group and the model group, which might have something to do with the pathogenesis of GAD.3The DXP could improve the ethology and the metabolism of relevant brain regions of GAD rats, which could tell that the anti-anxiety feature of DXP with the effect of soothing liver, clearing heat and tonifying spleen was probably related to its accommodation in relevant cerebral metabolism. This found completed the theory about treating GAD with Chinese medicine further more.
