[Background]PD (Parkinson’s Disease) is a kind of neurodegenerative diseases, as the main symptom of resting tremor, rigidity, slow movement, postural reflex disorder, and is progressive increased. With the progression of disease, it will serious impact the patients’quality of life. With in-depth study of the disease, more and more patients with early PD was diagnosed, PD is currently second only to Alzheimer’s disease, as the degenerative disease of the nervous system. Currently, the treatment of this disease is more focused on medication or surgical treatment, surgical treatment has some indications, is quite expensive; drug treatment can only improve symptoms, can not prevent the development of disease, and the symptoms of dyskinesia and fluctuations in motor complications become the more difficult issues during the treatment process. Chinese traditional medicine treatment has the characteristics of little toxic side effects, and can regulate yin and yang balance of the the patient through dialectical theory. Therefore, to further explore the advantages of Chinese medicine treatment of PD has important significance.Classic pathology change of PD is midbrain dopaminergic neuronal degeneration lost, and the residual neurons form into Lewy bodies --a kind of protein inclusion bodies, within which the major component is a-syn. a-syn (alpha-synuclein) was originally discovered in 1988 and is a kind of synaptic terminals of a precursor protein, its abnormal accumulation may be the cause of the death of dopaminergic neurons in PD patients. Its location in the brain is in the end of pre-synaptic and the neuronal nuclei. The recent clinical and basic studies suggest that PD pathology may be originated from the gastrointestinal ganglia, spinal cord, etc. and then gradually spread to the brain and other.[Purpose and Method]In order to further study the possible mechanism of traditional Chinese medicine treatment of PD, this subject is on the theory of "kidney is the collection of marrow", " kidney masters stool", "kidney masters practice", and will adopt the method of nourishing kidney and marrow, using animal models, and targeting with a-syn, to explore the mechanism of nourishing kidney and marrow fomular from the molecular level.Experimental study takes a-syn as a target, using rotenone, simulate the pathogenic process of "chronic low-dose oral cause stomach and intestinal ganglia, brain a-syn accumulation" in order to simulate the chronic multisystem progression of PD; by the observation of behavior and brain morphology in mice, to preliminarily explore modeling method in mice the behavior, morphology influence and efficacy of Chinese medicine treatment of PD. Immunohistochemical expression was observed for brain stomach, intestinal a-syn positive cells; using western blot, ELISA to compare the different content of a-syn in brain, stomach, intestine, in order to explore that how a-syn plays the role in the onset of PD, and its the relationship between PD models, and to explore the intervention mechanisms of nourishing kidney and marrow fomular of the brain, stomach, intestines dopamine neurons.[Experimental study]:Impact of nourishing kidney and marrow fomular on mice brain, stomach, intestines neurons a-syn Content[Objective] Using the observation of behavior, to preliminarily explore modeling method in mice behavior influence; using western blot, ELISA, immunohistochemical to compare the different content of a-syn in brain, stomach, intestine, in order to explore that how a-syn plays the role in the onset of PD, and efficacy of Chinese medicine treatment of PD.[Method] In this study, rotenone modeling, to simulate PD pathology and neurobiological changes from the PD pafhogenesis, the behavioral test is designed to verify successful modeling. Using western blot, ELISA, immunohistochemical to compare the different content of a-syn in brain, stomach, intestine, in order to explore the efficacy and possible mechanisms of Chinese medicine treatment of PD.[Results] 1, modeling rotenone group were in line with the pathogenic process of the "progressive disease development" in PD; 2, the model mice brain tissue is found a-syn positive cells increased and abnormal cytoplasmic a-syn aggregation, which a-syn protein concentrations were higher than the control group (P<0.05); after nourishing kidney and marrow fomular and Madopar therapy can improve these indicators; 3, model mice cells and the secretion of gastric glands appear at both a-syn positive cytoplasm of cells increased and abnormal a-syn aggregation, in which a-syn protein concentrations were higher than the control group (P<0.05), after nourishing kidney and marrow fomular and Madopar therapy can improve these indicators; 4, the model group intestinal endocrine cells and ganglion occurs at increased a-syn positive cells, nuclear stained. And a-syn optical density higher than the control group (P<0.05); after the Treatment of nourishing kidney and marrow fomular can improve these indicators.[Conclusion] Rotenone PD mice can produce behavioral change, and to produce PD symptoms in mice by enhancing the brain, stomach, intestine neurons a-syn content, Kidney Yisui only by reducing the mouse brain, stomach, intestine neurons a-syn content thereby improve symptoms.
