Effects Of 6'- Hydroxy - Jue - Ning Tablets And Curcumin On The Apoptosis Of HepG2 Cells

Objective:6’-hydroxyl Justicidin B belongs to lignans compounds and its antitumoreffect has not been reported.Curcumin has anti-inflammatory effect and can inhibitmalignant tumor growth progress,but its influence on apoptosis proteins like Bax andBcl-2is not very clear.We used MTT, western blot (WB) and other methods to studythe effect of6’-hydroxyl justicidin B and curcumin on liver cancer cell proliferationand bcl-2, bax protein expression, to explore the antitumor mechanism of twotraditional chinese medicines.Method:HepG2cells were cultured two days to make sure90%of the bottom of flaskwould be cells, and then,6’-hydroxyl Justicidin B or curcumin were given.MTT andWestern Blot were performed separately to test level of proliferation and expressionof Bax and Bcl-2of HepG2cells.Result：1.6’-hydroxyl Justicidin B can inhibit the proliferation of HepG2cells andshow the concentration-dependent manner (IC50is12μmol/L);the result of WesternBlot show that6’-hydroxyl Justicidin B can downregulate expression of anti-apoptosisproteins Bcl-2and upregulate the expression of apoptosis-promoting protein Bax.2.Curcumin can inhibit the proliferation of HepG2cells and show theconcentration-dependent manner (IC50is40μmol/L);the result of Western Blot showthat6’-hydroxyl Justicidin B can downregulate expression of anti-apoptosis proteinsBcl-2and upregulate the expression of apoptosis-promoting protein Bax.Conclusion： This study investigated the mechanism of HepG2apoptosis bysecondary metabolisms ofchinese medicine. The results showed that,6’-hydroxylJusticidin B can inhibit the proliferation of HepG2cells,which mechanism may be6’-hydroxyl Justicidin B can promote HepG2cells apoptosis through regulatingmitochondrial apoptosis protein.The mechanisms of action of curcumin on promotingHepG2cell apoptosis may be associated with downrgulatting the expression ofBcl-2protein and upregulatting the expression of Bax protein.
