A Case Of High Expression Of CyPA And Methotrexate Resistance And Case Of Thymoma

Objective:To exoplore the effect of exogenous overexpression of CyPA gene on the sensitizationto OXALIPLATIN induced apoptosis.Methods:1. CypA genes were respectively intuoduced into HCT116cells cells by liposome2000.Cellular CyPAin the transfected cells were assessd by western blot.2. MTT(4,5-dimethylth iazoleo r2,5-diphenylte trazoilumb romid,MTT)was adopted todetect the sensitivity of HCT116cells transfected with blank plasmid or targeting geneto OXALIPLATIN.3. FCM(flow cytometry) was used to detect the cell cycled is tribution and the rate ofapoptosis upon32ug/mL treatrment OXALIPLATIN for12h hours.PI staining was usedto detect apoptosis morphous of cells after treated with32ug/mL OXALIPLATIN for12hours.Rusults:1. Transform HCT116cells with recombinant plasmid pcmv-CyPA and blankplasmid,and detect instantaneously expressed product CyPAby western blot.2. Etopic expression of CyPA of HCT116-CyPA cells resulted in decreased proportionof G1stage,and increased proportion of S stage,than vector controls.3. The cell growth inhibition effects of OXALIPLATIN show that， the CyPAoverexpression groups showed higher resistance to OXALIPLATINL than the blankplasmid groups4. OXALIPLATINL induceded cell death via apoptosisin ducing effect.The apoptosisrates of blank plasmid groups and CyPA overexpression groups, after treated with32ug/ml OXALIPLATINL for12h were12.51%and6.31%. Conclusions:1. Ectopic overexpression of CyPAcould affects cell cycle tribution of HCT116.2. Ectopic overexpression of CyPAcould increase HCT116resistance to the cytotoxicityof OXALIPLATIN.
