Objective:To study the effect of hyperuricemia on atherosclerosis formation in rats.Methods:50male wistar rats were randomly divided into high fat diet group, high adenine diet group and normal control group. The high fat diet group rats were administered intragastrically with fat emulsion (5mL/kg/d), the adenine diet group were administered intragastrically with adenine (100mg/kg/d) and injected beneath the skin with oxonic Acid, and the control group was administered intragastrically with equal volume water. The level of serum UA, TC, TG, LDL and HDL were detected, and the pathological changes of rat aorta were observed. The expression of PCNA in vascular smooth muscle cells was detected by immunohistochemical analysis.The transcriptional changes of MCP-1,ICAM-1,VCAM-1were studied by RT-PCR.Results:Compared with the normal control group, the level of serum UA increased significantly in the high adenine diet group after4weeks (P<0.05),TC, HDL also increased (P<0.05),but LDL and TG were no significant change compared with the normal control group (P>0.05).The levels of TC, TG, HDL and LDL were significantly different in high fat diet group compared with the normal control group (P<0.05),_but the level of serum uric was no changed (P>0.05). As high adenine diet loaded for8weeks, the rat blood vessels appearred lamellar calcified plaque. As high fat diet high adenine diet loaded for12weeks, the deposition of calcium salt appearred diffusely in the middle and "medial" layer of the blood vessels. The expression of PCNA protein was increased (P<0.05) and MCP-1,ICAM-1,VCAM-1mRNA expression also increased in high fat diet group and high adenine diet group compared with the normal control group (P<0.05).The level of MCP-1mRNA transcription in high fat diet group was higher compared with high adenine diet group (P<0.05)Conclusion:Increasing serum uric acid can result in the form of atherosclerosis in rats.
