Objective: To investigate the role and significance of Th17ã€interleukin-21in thepathogenesis of pulmonary fibrosis.Methods:72SD rats were randomly divided into three groups: pulmonary fibrosis modelgroup (P, n=24), dexamethasone3ml/kg intervention (D,n=24), normal saline control group (N,n=24). GroupP, D injected Bleomycin (5mg/kg) intratracheal to establish rat model ofpulmonary fibrosis, Group N intratracheal instillation equal amount of0.9%sodium chlorideinjection. Group D injected dexamethasone intraperitoneal once a day from the7th date; GroupN, P injected of the same amount of0.9%sodium chloride injection.7,14,28days aftermodeling randomly sacrificed rats (8/group) to observe the pathological changes anddeterminate hydroxyproline of lung tissue; Determinate Th17cells accounted of proportion ofCD4+T cells by flow cytometry..Detect the expression level of IL-21by fluorescent RT-PCRmethod.Results: After Bleomycin injection, rats lung tissue inflammatory reaction earlier, and thegradual emergence of fibrosis and collagen fibers c. IL-21expression levels the the P group,Th17cells accounted the the proportion the CD4+T cells significantly higher than the the theN group (P <0.05), the highest expression levels at day7, then decreased gradually.Results: according to the lung tissue morphology, pathology observed, intratrachealinjection of bleomycin resulted in the preparation of pulmonary fibrosis successful. the lungtissue expression levels of IL-21and TH17cells accounted for the proportion of CD4+T cellsin P,D group were higher than N group, the difference was significant (p <0.05). the lungtissue expression levels of IL-21and TH17cells accounted for the proportion of CD4+T cellsin P group were higher compared with the D group, the difference was significant (p <0.05).Conclusion: Th17cells and IL-21factor plays an important role in the pathogenesis ofpulmonary fibrosis in rats, may be involved in the occurrence and development of pulmonaryfibrosis.
