This study was initiated to induce callus from shoots, roots, stems and leaves of Panax ginseng. The results showed different explants can all be induced to form callus and there was no obviously different in induced rate. Effects of 2.4-D, BA, NAA and KT on callus induction were investigated by using the orthogonal design.Statistic analysis indicated the optimal medium for callus induction is MS+2.4-D 4mg·L-1+ BA1.5mg·L-1.Selection of culture conditions was also studied. Effects of the type of medium. Carbon source, nitrogen source and different growth regulars on callus growth. Saponin content and saponin yield were investigated. We conducted the concentration of source is 3%, the concentration of NH4NO3 is 1650mg·L-1 and the concentration of 2.4-D, BA are 4mg·L-1. 0.5mg·L-1 respectively. In this condition ,the callus have high growth rate and saponin content and saponin yield are also improved significantly. The inoculum quantity, value of medium . temperature and light were deeply studied. 22℃, dark, pH6.0 of medium and inoculum quantity of 2-4g/fiask are the best condition for callus culture. In addition, MS+ BA 1mg·L-1+ KT 1.5mg·L-1 is the best medium for shoot propagation. The study investigated the influence of the salicylic acid elicitor on the gynthesis of saponin incallus culture. The result showed that salicylic acid enhanced greatly the accumulation of saponin. But the elicitation dosage and addition time brought different influence for saponin content. The saponin yield and saponin content were significantly increased in the presence of 10-3 mg·L-1 salicylic acid on day 28 and the maximum content of them were 34%. 64%higher than untreated callus respectively. Whereas polysaccharides, peroxidase ,phenylalanine ammonilyase,polyphenol oxidase were 50.5%,54%,6.1%,5.6%higher than untreated callus.
