It's one of important fields in biotechnological research to carry out gene-engeneering breeding of trees. As one of main sylviicultural trees, populus is very important in ecology and application, and has especial trait in scientific research, sos gene family is an important gene family that is correlated with salt tolerance, whether sos gene expressed highly in trees has great significance in science and foreground in application. In this study, the sosl and sos2 genes were introduced into eleven poplars, such as Populus tomentosa Carr, P.alba × brolinensis cv. Yinzhong and P. ussuriensis Kom., by Agrobacterium-mediated method, Agrobacterium-mediated in vivo and pollen-tube pathway. Transgenic plants were obtained finally and the function of the two genes was verified to study the value in application of sos gene introduced into trees.The effective transformation system of Agrobacterium-mediated method for Populus tomentosa Carr. was established .The time of infection and co-cultivation were important influencing factors in genetic transformation experiment by Agrobacterium-mediated method, the optimal infecting time was 15 minutes, and the co-cultivating time was 2 days. The concentration of antibiotic was also confirmed, 50mg/L was optimal for selection of resistant plants. The effective transformation system through Agrobacterium-mediated pathway in vivo for eight poplars, such as P.alba×brolinensis cv. Yinzhong, was developed. The time of stem cutting and the age of poplars has great influence on regeneration of stresses, while the location of stem cutting had little influenceffect by Agrobacterium-mediated method in vivo.The optimal time of stem cutting was June and July, and the optimal age of poplar was 2 years old. The transformation system of pollen-tube pathway for P. nigra L. × P. Simonii Carr. and P. ussuriensis Kom. was optimized. The concentration of plasmid and the time for transformation were key factors to seed setting percentage. The optimal concentration of plasmid was 30ug/ml, transferring at 3 hours after blossoming was able to get high seed setting percentage.PCR testing to the Kan-resistant plants of Populus tomentosa Carr. obtained by...
