| To breed benatazon sensitive male sterility lines utilizing the bel gene for sensitivity to herbicide benatazon and use them in hybrid seed production can help to prevent the contamination caused by selfing of male sterility lines effectively. Based on the systematical study on frequency, characteristic and distribution of SNPs in bel gene region, SNPs linkage map of the bel gene region were constructed and bel gene was fine-scale mapped in order to provide necessary basis for breeding benatazon sensitive male sterility lines with bel gene by marker-assisted selection. The main results are as follows:1. 17.6 kb DNA fragments of 24 loci in 24 different rice accessions were sequenced and 96 SNPs were found with SNPs frequency (θ) equaling to 1.31×10-3 and (π) equaling to 1.81×10-3. 23 SNPs was found in 5.7kb coding region with SNPs frequency (θ) equaling to 1.19×10-3and (π) equaling to 1.57×10-3 and 63 SNPs were found in noncoding region with SNPs frequency (θ) equaling to 1.37×10-3 and (π) equaling to 1.91×10-3. In the 24 loci surveryed, SNPs were found in 22 loci and none were found in only 2 loci.2.11 SNPs were selected base on the above results to construct SNPs linkage map of the region of bel gene. It was showed that the bel gene was located within the 92kb region between two SNP markers (SNP158 and SNP138), and SNP161 was cosegregated with the bel gene.3. Comparison of the DNA sequence between the mutation 8077s and wild W6154 indicated that a deletion of a single nucleotide in the coding region of cytochrome P450 hydroxylase gene on rice chromosome 3 might be the direct cause of the sensitivity to bentazon in rice.In present study, it was suggested that SNPs is of supreme use in the fine-scale mapping of genes and the strategy and method of breeding benatazon sensitive male sterility lines with bel gene by marker assisted selection were proposed.
