Actinobacillus Pleuropneumoniae is the causative agent of Porcine Pleuropneumoniae, a worldwide disease causing tremendous economic loss to the swine industry. It is a common infectious disease which infects the respiratory tract of pigs. The symptoms of infected pigs induced depression, difficult respiratory and pneumonia. With the development of the raising cosmically, Porcine Pleuropneumoniae becomes one of the five primary disease that threaten mordern pig industry.The following four parts were included in the studies:PartⅠ: Preparation, Application and Serotype of Outer membrane proteins on Actinobacillus pleuropneumoniaeThe outer membrane proteins of serotype 1-12 to Actinobacillus pleuropneumoniae were collected and roughly purified, then the extracted outer membrane proteins were serotyped. And then, the extracted outer membrane proteins were used as antigens to challenge the mouse with the same serotype bacteria, the results showed that the mouse were well protected immunized with 16μg outer membrane proteins. The mouse challenged with the same serotype bacteria were not dead at this dose , while the mouse of control group were all dead. Two days after immunization, IgG was examined in the mouse. At the 6th day, the mouse exhibited significantly high IgG against outer membrane proteins. Then the IgG slowly began to decline. In this research we found that there were no cross-reactions among serotype 3, 4 and 5, most of the mouse challenged with the different serotype bacteria were dead. Other serotypes showed apparent cross-reactions, no mouse were dead after challenging with the different serotype bacteria. These results revealed that outer membrane proteins would be one of the main protactive antigens of Actinobacillus pleuropneumoniae.
