| Actinobacillus Pleuropneumoniae is the causative agent of Porcine Pleuropneumoniae, a worldwide disease causing tremendous economic loss to the swine industry. It is a common infectious disease which infects the respiratory tract of pigs. The symptoms of infected pigs induced depression, difficult respiratory and pneumonia. With the development of the raising cosmically, Porcine Pleuropneumoniae becomes one of the five primary disease that threaten modern pig industry.The lipopolysaccharide of serotype 3, 4, 5, 7 and 8 on Actinobacillus pleuropneumoniae were collected and roughly purified by hydroxybenzene - water, then the extracted lipopolysaccharide were used as antigens to challenge the mouse with the same serotype bacteria, the results showed that the mouse were well protected immunized with 20μg lipopolysaccharide. The mouse challenged with the same serotype bacteria were not dead at this dose , while the mouse of controled group were all dead. These results revealed that lipopolysaccharide would be one of the main protective antigens of Actinobacillus pleuropneumoniae. This research provided the foundation for preparation and application of the subunit vaccine on Actinobacillus pleuropneumoniae.The polystyrene microtitration plates was coated with the purified lipopolysaccharide to Actinobacillus pleuropneumoniae of serotype 5 and serotype 7. The plates should be kept a whole evening at 4℃after an hour at 37℃. Then we blocked the plates, joining the sera and the antibody conjugated with HRP, and the steps above were done at 37℃for an hour.
