Wheat gluten (glutenin) subunits include high molecular weight glutenin subunits (HMW-GS) and low molecular weight subunits (LMW-GS), and HMW-GS relate to the bread properties closely. The identification of HMW-GS and the HMW-GS composition analysis mainly depends on the differences between HMW-GS electrophoresis migration at present. But the migration is not always identical to the HMW-GS, so it is difficult for HMW-GS analysis and the same wheat material sometimes has different analysis results. Moreover, this method is based on the seeds ruination, timewasting and painstaking, so this method is limited in screening and identification of lots of hybrid progenies, while AS (allelic specific) -PCR markers system can derecterly identify the composition of HMW-GS.The meathod is not limited by the growing-time and ensure the accuracy of subunits identification. In this stuy, according to our realities, the single and duplex AS-PCR system for idenfication of subunit gene with good bread-making quality of Glu-A1 and Glu-D1 were established. The method can be an efficient alternative to the standard separation procedure for early selectiom of usefule wheat genetypes. Addedly, the quantitative trait loci (QTLs) of 7 quality traits were mapped using a population of recombinant inbred lines (RILs). These QTLs might be useful for quality molecular marker assited selection in grain hardness and lipid straits. The main results are as follows:1.
