Barley is a typical self-pollinate crop, rich of types, distributed throughout the world. Its characteristics are easily to change after treated with the mutagensis, so it is an ideal experimental crop in scientific research.Antisense phospholipase Dγ(PLDγ) gene was introduced into Barley457 and Barley335 growing in the field by Agrobacterium Tumefaciens. The transformants were tested by PCR and PCR-Southern based on kanamycin selection. The results showed that anti-PLDγgene had been integrated into barley genomes. Meanwhile, the study of variance between the transgenic lines and the receptors in salt-resistance, drought-tolerance and agronomic characters were done, the major findings are as follows:1. Antisense phospholipase Dγ(PLDγ) gene was introduced into experimental materials via Agrobacterium tumefaciens. After rigorous Km -selection in the media, we obtained the Km-resistant plants. The 430bp special fragment of the plasmid was amplified in some Km-resistant plants by specific PCR, and the same conclusion in PCR-Southern, which showed that anti- PLDγgene was integrated into barley genomes.2.
