High-quality parents which had good HMW-GS such as Glu-A12*, Glu-B1x14+y15, Glu-D1x5+y10 and parents with high yield and disease resistance were crossed with Taigu-male-sterile wheat to establish a base population in this experiment. HMW-GS in recurrent selection population were screened rapidly by SDS-PAGE and novel PCR, and the selected seeds were sewed in the field and separated space.Meanwhile, genetics laws of HMW-GS in the population and some agronomic characters of C4 and C5 were also analyzed. Follows are the major results:1. New types of HMW-GS combination different from the parents such as"2*, 7+8, 5+10"and"1,14+15,5+10"etc. and many kinds of HMW-GS polymers such as"14+15,17+18","7+8,14+15","2+12,5+10"etc. were created in the population. The rare combination"1, 14+15,10"etc. were also created. All the above showed that using Taigu-male-sterile wheat recurrent selection is an effective way to improve the population's quality and create new subunits types.2. Similar trends of HMW-GS'genetic variance were found in both sterile and fertile population, which has more heterotypes in the early stage and varied widely, but with the further selection, its range becomes narrow. Composition of HMW-GS in the C2F1 fertile population varied widely, which has 10.0% heterotypes and 5.4 subunits each plant. However, in the C4F1 fertile population, no heterotypes were found and the major subunits were increased from 76.8% in the C2F1 fertile population to 94.8%.It means that when the selection come to the C4F1 the composition of HMW-GS in the population were nearly pure and genotypes of population were nearly homogeneity.
