Asparagus planting area in China is very large, but the seeds are supplyed by other countries completely at present, which increased the production cost of Asparagus greatly.Asparagus is dioecism in general. The yield of male plant is higher than the female plant. The subject of this paper is to establish the rapid propagating system super male plant of asparagus In Vitro, so that to produce a great deal of super male asparagus and to increase the asparagus yield.The results indicated that the method of killing germs is very important in the process of establishment of the pure culture and rapid propagation system. The explant should be immersed in 75% alcohol for 1 minute, and then washed with distilled water. After that, immersed in 0.1% Hgcl2 for 10 minutes, and then washed six times with distilled water.The optimum medium for shoot tip cultivation of asparagus is MS+6-BA 1.0mg/l + NAA 0.05 mg/ l.The optimum ratio of 6-BA to NAA for buds germination is 0.5:0.5. The average propagating coefficient could reach 2.5. In addition, the cultivating temperature influences the differentiation of tube seedling. The result showed that 27℃is optimum.It is showed that the RGR (roots germination rate) could reach 90% under the condition of 3 mm length bud and of 1/2 MS+IBA 0.5 mg/ l + KT 0.05 mg/ l.Root quality is an important factor in the domesticating process of asparagus tube seedling, because it affects LR (living rate) of in vitro plantlet significantly while transplanted from medium to soil. Transition culture can increase the LR enormously. Trained in the optimum sunlight and RH, transplanted to the disinfected planting media (soil 5: vermiculite 3), and kept in higher humidity for 30 days are also helpful for increasing LR of tube seedlings of asparagus.
