The first generation culture:1.1 Sterilization:To sterilize single bud stem with 75% alcohol and 2%NaClO solution, and a variety ofcombinations with unequal sterilization time were designed. The test shows that thecombination of 10s /7min has the best effect for eliminating bacterium.1.2 Selecting different partition of explant, the culture effect is evidently different.To select shoot tip, regular nod, laminae, and root segment from fragrant grape withoutkernel to perform a variety of cultures. The result shows that the shoot tip hasadvantage preponderance in regeneration inducement, and it is the best explant choicefor regeneration.1.3 The effect of growth regulator for the fist generation culture:The plant hormone is chosen for this test: cytokinin 6BA and plant auxin NAA, IBA,IAA. Among 10 combinations designed with reference to literature documentation andprevious test result, Ms+ 6BA0.5mg/L+ IAA0.5mg/L is the most suitable for shoot tipregeneration of fragrant grape without kernel.2. The effect of growth regulator for subculture:The further sieving was made for hormone formula in the subculture. Among 10 formulas,MS+6BA0.5 mg/L +IAA0.5 mg/L is the most suitable for "fragrant grape without kernel"subculture. Active carbon shows inhibiting effect for bud browning.3.
