| Fungal disease occurred very frequently on grape cultivars in recent years; general medicine control had great hazardness and localization, and the traditional breeding technique procured diseaseresistant race very hardness. But now, Biotechnology having large foreground in breeding means and the consummate organizational culture technology afforded powerful support for building virus free gene stock and transgene breed in grapevine. However, too hypo regeneration frequency restricted transgene project developing of grape now. Conquering this conundrum and improving regeneration became the hot spot research in grape biotechnology breeding. In the test, regeneration system was studied using the in vitro stem apex, leaf, petiole, stem and root of 'Red Globe', at the same time, discussed another seven Vitis vinfera L. grape regeneration —'jingxiu', 'GN-18', 'Autumn Royal', 'Superior seedless', 'Centennial seedless', 'Victoria'and 'Zana'. And studied the effect factor on regeneration such as medium type, hormone type and density, cultivate condition. At last, searched the cytology structure of grape adventitious bud. And obtained thereinafter results: 1. As to sieving the different mediums type, MS was confirmed the best suitable medium. Cultured 20 to 25 days test-tube plantlet and 10days dark foster at first, then giving about 1000Lx low irradiance could get tiptop regeneration rate. 2. On the feasible regrowth condition, leaf and stem apex can attain large cluster bud from In Vitro Grape Plantlets. 3.The leaf best hormone match of five varieties were: 'Red Globe'MS+ 6-BA1.5mg/L + IBA0.01mg/L; 'Superior seedless'MS + 6-BA1.5mg/L + IBA0.01mg/L ; 'GN-18'MS + 6-BA2.0mg/L + IBA0.01mg/L; 'Victoria'MS + 6-BA0.75mg/L + IBA0.01mg/L; 'Zana'MS + 6-BA2.0mg/L + IBA0.01mg/L.The best stem apex regeneration hormone match was 1/2MS + 6-BA1.5mg/L + NAA0.01mg/L + LH100mg/L. Best of all radication medium of adventitious bud was 1/2B5+IBA/IAA 0.5mg/L+sucrose 15g/L. Ranged from eight cultivars, three breeds of 'Centennial seedless', 'Autumn Royal'and 'jingxiu'couldn't creat adventitious bud, the other five's has well regrowth, thereinto, the 'Superior seedless'had best regeneration rate 17.4%, then 'Zana'14.28%, 'Victoria'12.3%, 'Red Globe'11.98%, 'GN-18'9.57%. 4. Pass observing the cytology of grape adventitious bud. We consider that the adventitious bud produced from principal vein of leaf came from split of nerve epidermis cell and vascular bundle thinwalled containers. Petiole anvil created adventitious bud caused from mesophyll tissue and vascular bundle thinwalled containers of leaf inside. Mesophyll cell of leaf epidermis can single split into group meristematic cell, then more differentiation make it developed into adventitious bud on leaf margin. |
