With the improvement of the people's incoming level, consumption of the beef is increasing rapidly, which represents the elevation of requirement to beef quantity. People are paying more attention to the beef quality. The beef quality is a very important economic trait. Because it can influences the persistent development of beef cattle industry and the economic benefit. It is indicated the Calcium-activated neutral protease 1 (Calpainl, CAPNl)gene is the candidate gene of influencing the beef quality. The reseach mainly screens the SNPs of calpainl gene and studys its influence to the beef tenderness, marbling, and acidity and meat color.With the bioinformatics methods and public database resource, six mutation loci of CAPN1 gene were validated, which is A3717G A3854G C4789T A5688G C5709G A6270G. The mutation: C5709G lead to the amino acid substitution of G (Gly) for A (Ala).All the ten populations is under the Hardy-Weinberg equilibrium condition in the six loci of CAPN 1 gene. And the G allele of A3717G locus is dominant gene in ten breeds. Hybrid cattle's genetic polymorphism is higher than purebred cattle, the Dachang Simmental hybrid's genetic polymorphism is the highest, the Hereford cattle is the lowest. The local cattle's genetic polymorphism is higher than the foreign cattle. For A3854G locus, most breed's genetic polymorphism is higher, except Luxi cattle and Hereford cattle. Allele T in C4789T locus, allele G in A5688G loucs, allele G in C5709G locus and allele G in A6270G locus are dominant allele genes. And the genetic characteristic of this four loci are very similar, and the genetic polymorphism are very lower. For A3854G locus, Luxi cattle's and Hereford cattle's genotype distribution are significantly different with others, genotype distribution of other loci is identical.In the ten populations,C4789T, A5688G , C5709G and A6270G is under the linkage disequilibrium, 16 halotypes was tested in CAPN1 gene by halotype analysis, A-A-T-G-G-G G-A-C-A-C-A G-A-T-G-G-G G-G-T-G-G-G is common, and others is fewer. The four loci is tight linkage.For A3717G locus, AG genotypes' and GG genotype's tenderness value are better than AA genotype's. For A3854G locus, GG genotype's tenderness value is significantly better than AA genotype's. But for C5709G locus there are no differences in three genotypes. The three genotypes of A3717G and A3854G loci have no difference for meat color. ForC5709G locus, the genotype means of CC is better than GC and GG (PO.05).The genotypes of each locus in CAPNl gene have no significant difference for beef marbling and beef acidity.For each locus in CAPNl gene, the marbling of Luxi cattle, Jinnan cattle and Qinchuan cattle is lower than others. The WBS of Angus cattle, Hereford cattle and Baolongshan Simmental cattle is lower, but the tenderness is preferable. The acidity of Angus hybrid cattle, Jinnan cattle, Limousin hybrid cattle, Qinchuan cattle and Hereford cattle is lower, meat quality is preferable. For Angus hybrid cattle, Dachang hybrid cattle, Charolais hybrid cattle, Baolongshan Simmental cattle and Angus cattle, Means of meat color is lower and the meat color is preferable;the marbling of cattle whose age betweentwo to three years is lower than others.
