It is very necessary for us to study the biological characteristic that the sunflower was a burgeoning oil plant and the potential of generalizing the soil areas of sunflower are very huge. With the developing of science, people will realize its advantages and uses increasingly, at the same time the abundant nutrition ingredients will be familiar with the people's life. Such as the sunflower oil is the main raw materials of dishing, cooking and processing delicatessens. The unsaturated fatty acid of sunflower oil is far higher than that of soja, colza oil and peanut oil etc. So it owns the important characteristic of dissolving cholesterins, it can be propitious to reducing the contents of cholesterin and preventing and curing haemal diseases and then building our body, prolonging the people's life.In the twenty years of last century, with the researching of aberrance breeding deeply, the physical and chemical means were used to causing mutation on sunflower and to breeding the new breed. Among all of these workings, Guan zhenqian of Shanxi Agriculture university used the neutron 5 × 1010n/cm to treat dry black sunflower seeds and breed the very short plant Jinkui No.1, but the work of inducing chemical mutation was nearly untouched. Foreign scholars used the liquor of dimethyl sulphate and hypo ethylamine to treat with the sunflower seeds and cultivate the sunflower varieties of being propitious to plant thickly. Internal in Changchun University of Agriculture and Animal Sciences, Ji jing and Wang ping induced the mutation of the sunflower seeds by the diethyl sulphate to discuss the mutation effects on sunflower, as a result it can be offered the evidences for developing the mutation breeding works in the future.Laser is used to treat the sunflower seeds, induce mutation, synchronously combine the breeding work, filter the excellent mutation plants, alter its bad characters, and then will satisfy the demands of agriculture production. Since the long biology evolution and natural selection, the species have formed the ability of adapting the nature strongly. Even if had been caused mutation, the species eliminated the bad individuals and reduced mutations with the individuals growing and evolving. Aiming at these facters, Sensitizing agent can be used to treat laterly and prevent from renewing the damage of DNA, as a result, the rate of mutation will be improve greatly. It has been proved that EDTA was a very effective sensitizing agent and increased the chromosome aberration evidently. Acyl compound such as acrylamide has been used to treat after eradiating and proved a very effective sensitizing agent also.In this text, Nd:YVO4 laser was used to treat the sunflower seeds and sensitizing agent lately, then observed and registered the rate of bourgeon, the rate of chromosome aberration of root tip, mitotic indexand the length of radicel to discuss the dosage-response Nd:YVO4 laser availing sunflower and the cell effect of sensitizing agent on laser irradiating the sunflower seeds, it could be established theoretic for after laser irradiating breeding.Because of these, the following aspects were studied:1. Nd: YVO4 laser was used with apparatus of output power 3mM> wavelength 532nnu irradiating dosage 338mW/cm2 to treat the sunflower seeds, the time were 0, 1 mhu 5mim 10min> 15mim 20min seperately. The result proved: Nd: YVO4 laser irradiated evidently chromosome aberration of the sunflower seeds, the rate of chromosome aberration was increasing with the adding time of laser treatment; the mitotic index decreased, the cell mitosis restrained, the length of radicel lessened. Of all these treatments, the most effective treatment was the 20 min of laser treatment.2. The sensitizing agent EDTA (lm mol/L) and acryl amide (5m mol/L) were used to treat the sunflower seeds separately, the time was 3 Ik 5h and 7h. It were found that all the three treatments irradiated the chromosome aberration, the rate of chromosome aberration was increased with the treatment time, the style of chromosome aberration was main the bridges; the mitotic index and the length of radicel decreased with the time treatment. The effect of aberration was the highest in 7h time treatment. So the scientific research works should consider the effect of aberration when used the sensitizing agent.3. Treated with Nd: YVO4 laser firstly, the time was 5 min> 10 min and 15 min, then used the EDTA and acrylamide lately, time was 3h> 5h and 7h.The results were that the rate of chromosome aberration of complex treatment was higher evidently than that of the laser treatment solely, the mitotic index and the length of radicel were decreased with the time treatment.4. Observed the effects of chromosome aberration of bourgeoning seeds treated by Nd: YVO4 laser, at the same time, compared with that of dry seeds. The results proved that the effects of chromosome aberration of dry seeds were higher than that of bourgeoning seeds. |
