| The leaves and leafstalks were used as explants to establish the technique system of tissue culture and to study the effects of different carbon sources in Oxalis triangularis Subeg. Triang. With orthogonal design, analysis of variance and multiple comparisons, the best medium compositions and the culture process were analyzed . Besides, the effects of sucrose,glucose and soluble starch on plant regeneration, colour and flowering were also investigated. The results were as follows:(1)MS+6-BA0.5mg/L+ NAA0.5mg/L+IBA0.1 mg/L gave rise to the good result in inducing adventitious buds from leaf explants, in which the inducement rate could be 90%.(2)MS+KT 1.0mg/L+NAA0.5 mg/L+IBA0.5 mg/L was the best medium in inducing adventitious buds from leafstalk explants where the inducement rate could be 93.33%.(3)MS supplemented with 6-BA1.5mg/L and 0.2mg/L NAA was favorable to the proliferation of buds ,in which the multiplication rate could be 4.44.(4)The growth of roots was best on 1/2MS medium added NAA 0.2mg/L ,Where the rooting rate could be 95%.(5)Sucrose was the best carbon source in regenerating plantlets from leaf explants 3% was the best sucrose concentration in which the plant regeneration rate was high and the plantlets grew well.(6)5% sucrose concentration was effective in inducing floral buds .(7)Alternate experiment between the medium with sucrose(3%) and themedium without sucrose showed that sucrose was the important factor determining the color of leaves of the plants. In the medium without sucrose the leaf colour changed from purple to green .After transferred to the medium with sucrose(3%),the leaf colour changed from green to purple .Sucrose had more effect on the content of anthocyan than that of chlorophyll .After the normal plantlets with purple leaves obtained on the medium containing 3% sucrose were transferred to the medium without sucrose the content of anthocyan and chlorophyll decreased gradually as time passed, but the reduction range of anthocyan was larger than that of chlorophyll. However when transferred to the medium with sucrose(3%) again the two pigments rose gradually, but the rising range of anthocyan was larger than that of chlorophyll. |
