| Sichuan-white geese and Landes geese were studied in this experiment. The influence on the composition of body and plasma parameters by force-feeding were analized ,and genetic difference between the two breed geese were analyzed as well with RT-PCR method . The expression characteristic of microsomal triglyceride transfer protein (MTP) gene in geese were studied. The expression pattern of MTP gene in geese liver and small intestine during the different growth stages were analized. The influence of force-feeding on the expression of MTP gene in geese liver and small intestine were showed. These results showed:The amount of feed was fed according to the body weight .The body weight, the liver weight and the percentage of lipid in liver of the breed geese significantly raised up after the force-feeding. But the percentage of water in the liver of the breed geese significantly went down after the force-feeding.The body weight, the liver weight ,the percentage of the liver weight , the subcutaneous adipose tissue weight and the percentage of liver lipid of Landes geese were significantly or very significantly higher than those of the Sichuan-whites geese, while the percentage of water in liver was lower than that of Sichuan-whites geese. The concentration of TG, VLDL and Insulin raised after force-feeding and the concentration of Insulin of Landes geese was very significantly lower than that of Sichuan-white geese. The concentration of Insulin in Landes geese and Sichuan-whites geese were 10.28 ± 0.31mmol/l and11.53±0.28mmol/l respectively, and the difference between these two breed geese was significant.After the force-feeding, the correlation between the liver weight and TG and VLDL, and between the subcutaneous adipose tissue weight, the abdominal adipose tissue weight and the body weight showed that there was the difference between Landes geese and Sichuan-whites geese and that the liver weight and the concentration of TG and VLDL of Sichuan-whites geese were highly correlated and the coefficient was r=0.68, PO.05 and r=0.55, P<0.05 respectively. While the Landes geese showed the feeble correlation and the coefficient was r=0.26and 1- - 0.14 respectively. The subcutaneous adipose tissue weight, the abdominal adipose tissue weight and body weight showed the moderate positively correlation in Sichuan-whites ; ,.ese, but the Landes geese showed no the positively correlation, which suggested Sichuan-whites geese had a strong lipid deposition in subcutaneous adipose tissue and the abdominal adipose tissue compared to Landes geese.The MTP gene of geese was expressed in liver and small intestine, and existed in brain at lOweek age. The expression pattern of MTP gene during the different period were that the expression of MTP gene was not detected in liver but detected in small intestine at 7days age. The expression abundance of MTP gene of Landes geese and Sichuan-whites geese in intestine was 0.72and 0.65 respectively, the amount of expression of MTP gene in liver and small intestine showed the raising trend from 10 week age to 16 week age . The expression abundance of MTP gene in intestine was higher than that of liver from 1 week age to 13 week age , and lower than that of liver from 13 week age to 16 week age .The effect of the force-feeding had the different influence on the expression of MTP gene in geese liver and small intestine: the expression lever of MTP gene in liver reduced while the small intestine increased after force-feeding . |
